The two functional domains of a cloned human fibroblast NADPH-cytochrome P450 reductase have been expressed in Escherichia coli and purified on the milligram scale for crystallization studies. One domain contains the cofactor FMN-binding site and the other contains the binding sites for cofactor FAD and substrate NADPH. Crystals of both domains have been obtained by the microbatch method. The crystals of the FMN domain belong to the monoclinic space group P21, with unit cell dimensions of a = 39.3 A, b = 51.5 A, c = 47.8 A, and beta = 105.7 degrees and have one molecule in the asymmetric unit. Diffraction data up to 2.3 A were collected with a merging residual on intensity of 9.3%. The crystals of the FAD/NADPH domain belong to the ortho-rhombic space group P212121 with unit cell dimensions of a = 55.9 A, b = 58.6 A, c = 131.1 A and have one molecule in the asymmetric unit. Diffraction data up to 2.6 A were collected with a merging residual on intensity of 8.0%.

This study is the first to use synthetic biological omics technology to analyze the molecular mechanism underlying deep degradation of TNT, to construct an artificial transformation system to create engineered Escherichia coli bacteria, and to use Bacillus subtilis as an expression host to explore the mechanism driving the reshaping of the deep degradation platform on microecology. Nitroreductase family protein, 2-oxoacid:acceptor oxidoreductase, NADPH-cytochrome P450 reductase, monooxygenase, ring-cleaving dioxygenase, and RraA family protein significantly participated in the reduction-hydroxylation-ring opening cleavage of TNT, achieving deep transformation of TNT to produce pyruvic acid and other products that entered the cellular metabolic cycle. The key toxic metabolic pathways of TNT, 2,4-diamino-6-nitrotoluene, 2,4,6-triaminotoluene, and 2,4,6-trihydroxytoluene are pantothenate and CoA biosynthesis.

Ferroptosis, a type of programmed cell death, represents a distinct paradigm in cell biology. It is characterized by the iron-dependent accumulation of reactive oxygen species, which induce lipid peroxidation (LPO), and is orchestrated by the interplay between iron, lipid peroxides, and glutathione. In this review, we emphasize the frequently overlooked role of iron in LPO beyond the classical iron-driven Fenton reaction in several crucial processes that regulate cellular iron homeostasis, including iron intake and export as well as ferritinophagy, and the emerging roles of endoplasmic reticulum-resident flavoprotein oxidoreductases, especially P450 oxidoreductases, in modulating LPO.

Human exposure to polycyclic aromatic hydrocarbons (PAH) is a significant public health problem that will worsen with a warming climate and increased large-scale wildfires. Here, we characterize an epigenetic memory at the cytochrome P450 1 A (CYP1A) gene in wild Fundulus heteroclitus that have adapted to chronic, extreme PAH pollution. In wild-type fish, CYP1A is highly induced by PAH.
