Nucleotide and deduced amino acid sequences of the outer capsid proteins VP4 and VP7 of a murine rotavirus strain (YR-1) isolated in Japan were determined. Comparisons of the VP7 amino acid sequence of YR-1 with other murine rotavirus strains (EB, EW, EC, EL and EHP) (1) showed that YR-1 was highly homologous to EB, EW, EC, EL and EHP. Moreover, YR-1 was more closely related to strains representing G3 than to any other G type. Analysis of the VP4 amino acid sequence revealed that YR-1 was highly homologous to EB, EW, EC and EL [tentatively P17 (1)], and more closely related to EHP [tentatively P18 (1)] than to any other P type. Enzyme immunoassay with monoclonal antibodies against G types (KU-4 and BH49 for G1, S2-2G10 and BW36 for G2, YO-1E2 and BC5 for G3; and ST-2G7 and BE18 for G4) and against a P type (YO-1S3, KU-12H and YO-2C2 for P8) showed no reactivity. These results indicate that YR-1 is highly homologous to EB, EW, EC and EL.

Bacterial flagellin, a potent intestinal innate immune activator, prevents murine rotavirus (RV) infection independent of adaptive immunity and interferons. The flagellin-induced immunity is mediated by Toll-like receptor (TLR5) and Nod-like receptor C4 (NLRC4), which elicit the production of interleukins 22 (IL-22) and IL-18, respectively. Here, we assessed whether a high abundance of flagellin at the time of vaccination would negatively affect the oral RV vaccine take.

Viruses engage in a variety of processes to subvert host defenses and create an environment amenable to replication. Here, using rotavirus as a prototype, we show that calcium conductance out of the endoplasmic reticulum by the virus encoded ion channel, , induces intercellular calcium waves that extend beyond the infected cell and contribute to pathogenesis. Viruses that lack the ability to induce this signaling show diminished viral shedding and attenuated disease in a mouse model of rotavirus diarrhea.

Porcine rotavirus A (RVA) is one of the major etiological agents of diarrhea in piglets and constitutes a significant threat to the swine industry. A molecular epidemiological investigation was conducted on 2422 diarrhea samples from Chinese pig farms to enhance our understanding of the molecular epidemiology and evolutionary diversity of RVA. The findings revealed an average RVA positivity rate of 42% (943/2422), and the study included data from 26 provinces, primarily in the eastern, southern and southwestern regions.

Enteric pathogen rotavirus (RV) primarily infects mature enterocytes at the tips of the intestinal villi; however, the role of secretory Paneth and goblet cells in RV pathogenesis remains unappreciated. Atoh1 knockout mice (Atoh1cKO) were used to conditionally delete Paneth, goblet, and enteroendocrine cells in the epithelium to investigate the role of secretory cells in RV infection. Unexpectedly, the number of infected enterocytes and the amount of RV shedding in the stool were greatly decreased following secretory cell deletion.
