It was shown that a suspension culture of Macleaya cordata accumulated benzo(c)phenanthridine alkaloids in the cell biomass. By the composition of the alkaloids and their content the cell culture markedly differed from the intact plant and callus tissue. The benzo(c)phenanthridine compounds in the cell culture were mainly represented by the reduced forms: dihydrosanquinarin and dihydrochelirubin. In the green plant they were mainly represented by the oxidized forms: sanquinarin and chelerithrin. The cell culture practically contained no chelerithrin characteristic of the intact plant.

Trigger valves are fundamental features in capillary-driven microfluidic systems that stop fluid at an abrupt geometric expansion and release fluid when there is flow in an orthogonal channel connected to the valve. The concept was originally demonstrated in closed-channel capillary circuits. We show here that trigger valves can be successfully implemented in open channels.

Mesenchymal stromal/stem cells (MSCs) are promising candidates for regenerative medicine owing to their self-renewal properties, multilineage differentiation, immunomodulatory effects, and angiogenic potential. MSC spheroids fabricated by 3D culture have recently shown enhanced therapeutic potential. MSC spheroids create a specialized niche with tight cell-cell and cell-extracellular matrix interactions, optimizing their cellular function by mimicking the in vivo environment.

Primary human mast cells (MC) obtained through culturing of blood-derived MC progenitors are the preferred model for the study of MRGPRX2- IgE-mediated MC activation. In order to assess the impact of culture conditions on functional MRGPRX2 expression, we cultured CD34-enriched PBMC from peripheral whole blood (PB) and buffy coat (BC) samples in MethoCult medium containing stem cell factor (SCF) and interleukin (IL)-3, modified through variations in seeding density and adding or withholding IL-6, IL-9 and fetal bovine serum (FBS). Functional expression of MRGPRX2 was assessed after 4 weeks via flow cytometry.
