The paper summarizes the hitherto knowledge concerning the absorption of perorally administered enzymes in the digestive tract. After the transport through the intestinal wall the enzymes are protected against immunity recognition and they reach the affected regions via blood and lymphatic routes. A survey of enzymes taken into consideration and the essence of their action are presented. In the final part, the fields are summed up in which systemic enzymotherapy proves useful.

Several clinical studies have shown that COVID-19 increases the systemic concentration of drugs in hospitalized patients with COVID-19. However, it is unclear how COVID-19-mediated bidirectional dysregulation of hepatic and pulmonary cytochrome P450 (CYP) 3A4 affects drug concentrations, especially in the lung tissue, which is most affected by the disease. Herein, physiologically based pharmacokinetic modeling was used to demonstrate the differences in systemic and pulmonary concentrations of 4 respiratory infectious disease drugs when CYP3A4 is concurrently downregulated in the liver and upregulated in the lung based on existing clinical data on COVID-19-CYP3A4 interactions at varying severity levels including outpatients, non-intensive care unit (ICU), and ICU patients.

During skin fibrosis, extracellular matrix (ECM) proteins are overproduced, and resident lipid-filled, mature dermal adipocytes are depleted in both human disease and mouse models. However, the mechanisms by which the reduction in lipid-filled adipocytes occurs during fibrosis are not well understood. Here, we identify that adipocyte lipolysis via the rate limiting enzyme, adipocyte triglyceride lipase (Atgl), is required for loss of adipose tissue during skin fibrosis in mice.

Aflatoxin B1 (AFB1) is a prevalent contaminant in food and feed matrices, known for its hepatotoxic effects. Its metabolic breakdown generates reactive oxygen species (ROS), leading to oxidative stress and subsequent liver damage. Mitigating oxidative stress is, therefore, essential for ameliorating the hepatocellular damage and systemic toxicity caused by AFB1.

Preventing bacterial infection and accelerating wound closure are critical for wound healing. Herein, a novel multifunctional polyvinyl alcohol-polyvinylpyrrolidone (PVA-PVP) microneedle (MN) patch embedded with enzyme-like activity (CoFe)(S) (CFS) nanoparticles and metal ions (Co and Fe) was systematically synthesized for the management of bacteria-infected wounds. CFS regulated redox homeostasis and achieved bacterial eradication while concomitantly alleviating oxidative damage.

Oligorecurrent prostate cancer (PCa) can be treated with metastasis-directed therapy (MDT), which may be performed using radioguided surgery (RGS) as an experimental approach. These procedures have shown promising outcomes, largely due to the high lesion detection rate of positron emission tomography/computed tomography (PET/CT).
