The rostral hypothalamus and adjacent basal forebrain participate in the generation of sleep, but the neuronal circuitry involved in this process remains poorly characterized. Immunocytochemistry was used to identify the FOS protein, an immediate-early gene product, in a group of ventrolateral preoptic neurons that is specifically activated during sleep. The retrograde tracer cholera toxin B, in combination with FOS immunocytochemistry, was used to show that sleep-activated ventrolateral preoptic neurons innervate the tuberomammillary nucleus, a posterior hypothalamic cell group thought to participate in the modulation of arousal. This monosynaptic pathway in the hypothalamus may play a key role in determining sleep-wake states.

Aggression and mating of male mice are strongly associated with Esr1-expressing neurons in the bed nucleus of the stria terminalis (BNSTpr) and hypothalamus in the vomeronasal pathway. By projecting to the downstream hypothalamus, the upstream BNSTpr gates mating and aggression of male mice and maternal behavior of female mice. The medial preoptic area (MPOA) and ventrolateral subdivision of the ventromedial hypothalamus (VMHvl) are two subdivisions of the hypothalamus downstream.

There are well-documented gender differences in the risk and severity of sleep disorders and associated comorbidities. While fundamental sex differences in sleep regulatory mechanisms may contribute to gender disparities, biological responses to sleep loss and stress may underlie many of the risks for sleep disorders in women and men. Some of these sex differences appear to be dependent on sex chromosome complement (XX or XY) and the organizational effects of reproductive hormones.
