In chloroplasts of plants the xanthophyll cycle is suggested to function as a protection mechanism against photodamage. Two enzymes catalyze this cycle. One of them, violaxanthin de-epoxidase, transforms violaxanthin (Vio) to zeaxanthin (Zea) via antheraxanthin (Anth) and is bound to the lumenal surface of the thylakoid vesicles, when being in its active state. The other enzyme, Zea-epoxidase, is responsible for the backward reaction (Zea-->Anth-->Vio) and is active at the stromal side of the thylakoid. For the epoxidation of Zea this enzyme requires NAD(P)H and O2 as cosubstrates. Using isolated thylakoid membranes we found that FAD enhances the epoxidase activity (decrease of apparent Km for NAD(P)H and two-fold increase of Vmax). The flavin functions as a third cofactor which is partially lost during the isolation procedure of thylakoids. Other flavins, such as FMN or riboflavin are without effect. The involvement of FAD in the enzymatic reaction is also demonstrated by the inhibitory action of diphenyleneiodoniumchloride (DPI) (IC50 = 2.3 microM), a compound that blocks the reoxidation of reduced flavins within enzymes. The Zea-epoxidase is a multi-component enzyme system which can be classified as FAD-containing, NAD(P)H- and O2-dependent monooxygenase that is able to epoxidize 3-hydroxy beta-ionone rings of xanthophylls in the 5,6 position.

Glucose deprivation, a hallmark of the tumor microenvironment, compels tumor cells to seek alternative energy sources for survival and growth. Here, we show that glucose deprivation upregulates the expression of mitochondrial-cytochrome c oxidase II (MT-CO2), a subunit essential for the respiratory chain complex IV, in facilitating glutaminolysis and sustaining tumor cell survival. Mechanistically, glucose deprivation activates Ras signaling to enhance MT-CO2 transcription and inhibits IGF2BP3, an RNA-binding protein, to stabilize MT-CO2 mRNA.

Genome-wide CRISPR screens are an effective discovery tool for genes that underlie diverse cellular mechanisms that can be scored through cell fitness. Loss-of-function screens are particularly challenging compared to gain-of-function because of the limited dynamic range of decreased sgRNA sequence detection. Phytohormone auxin plays a pivotal role in governing plant growth, development, and responses to abiotic stresses. YUCCA6 (YUC6), an auxin biosynthetic enzyme belonging to the flavin monooxygenase (FMO) subfamily, converts indole-3-pyruvic acid to indole-3-acetic acid. Our prior investigation uncovered that YUC6 also functions as a thiol-reductase and chaperone in a Cys85-dependent manner, resulting in conferred tolerance to nickel heavy metal stress and drought and delayed leaf senescence.

Research into addictive eating has gained traction over recent years, however there is still debate among experts surrounding the nature of the condition. Using reflexive thematic analysis this paper considers addictive eating through a participant focused lens, specifically focussing on the phenomena of the food environment and its impact on individuals with addictive eating. Semi-structured interviews were undertaken to explore the participants' ( = 10) perspectives with addictive eating.
