An express method has been elaborated for identification of human and bovine species mycobacteria in the polymerase chain reaction. The high specificity of the technique and simplicity of material preparation for research make the experimental procedure simple and quick, permitting one to identify Mycobacterium tuberculosis and Mycobacterium bovis directly in pathogenic material.

This study investigated the diagnostic efficiencies of two assays for the detection of complex: (1) the reciprocal-flow real-time polymerase chain reaction (PCR)-based GeneSoC assay and (2) the real-time PCR based GENECUBE MTB assay with quenching probe. These assays were performed for stored clinical samples and results were compared with the confirmed results based on culture and COBAS TaqMan MTB assay. A total of 53 samples (20 confirmed positives and 33 confirmed negatives) were included in the performance analysis.

Infectious complications in peritoneal dialysis (PD) remain a constant challenge, with atypical pathogens posing significant risks. This case from Thailand highlights the rare occurrence of , an often-overlooked non-tuberculous mycobacterium (NTM), as the causative agent in a catheter-related exit-site infection that progressed to peritonitis. Initially misattributed to from preceding exit-site infections, was ultimately identified as the primary pathogen through multiple effluent cultures and advance polymerase chain reaction sequencing.

Hepatic tuberculosis, typically associated with miliary tuberculosis, can occasionally present as localized liver lesions. This case report describes a 77-year-old male presenting with persistent abdominal pain and fever, following an endoscopic retrograde cholangiopancreatography for bile duct sludge removal. Subsequent computed tomography revealed focal liver lesions.

It was a general belief that drug resistance in Mycobacterium tuberculosis (Mtb) was associated with lesser virulence, particularly rifampicin resistance, which is usually produced by mutations in the RNA polymerase Beta subunit (RpoB). Interestingly, this kind of bacterial mutations affect gene transcription with significant effects on bacterial physiology and metabolism, affecting also the bacterial antigenic constitution that in consequence can produce diverse immune responses and disease outcome. In the present study, we show the results of the Mtb clinical isolate A96, which is resistant to rifampicin and when used to infect BALB/c mice showed hypervirulence, apparently by rapidly polarization of the Th2 immune response through early and high production of IL-4.

Rifampicin-resistant tuberculosis (RR-TB) is a critical issue with significant implications for patient care, public health, and TB control efforts that necessitate comprehensive strategies for detection. This study presents a novel point-of-care diagnostic tool for RR-TB detection employing a peptide nucleic acid (PNA)-paper-based sensor combined with isothermal recombinase polymerase amplification (RPA). The sensor targets mutations in codons 516, 526, and 531 of the rpoB gene, the top three common mutations associated with rifampicin-resistant strains.
