Thymic myoid cells produced macrophage lineage cell stimulatory factors. Activities were separated into two factors on DEAE-Sepharose CL-6B chromatography: one eluted at lower concentrations of NaCl and the other at higher concentrations of NaCl. The latter fraction was purified to homogeneity with an apparent molecular weight of 100,000. This factor stimulated the growth of macrophage-lineage cells from the bone marrow, but not that of granulocytes, megakaryocytes or erythroblasts. The 100,000 MW factor was able to induce Ia antigens on proliferating bone marrow cells. These results suggest that myoid cell-derived 100,000 MW factor plays significant roles in the generation of Ia-positive macrophage lineage cells which are important for T-cell development in the thymus.

Hereditary transthyretin amyloidosis with polyneuropathy (ATTRv-PN) is a neurodegenerative disease caused by mutations in the gene encoding transthyretin (TTR). Despite amyloid deposition being pathognomonic for diagnosis, this pathology in nervous tissues cannot fully account for nerve degeneration, implying additional pathophysiology for neurodegeneration, which, however, has not yet been fully elucidated. In this study, neuroinflammation in ATTRv-PN was investigated by examining nerve morphometry, the blood-nerve barrier, and macrophage infiltration in the sural nerves of ATTRv-PN patients and the sciatic nerves of a complementary mouse system, i.

Background: Bone marrow inflammaging is a low-grade chronic inflammation that induces bone marrow aging. Multiple age-related and inflammatory diseases involve bone marrow inflammaging. Whether common pathological pathways exist in bone marrow inflammaging remains unclear.

We have previously identified that infection induces a unique form of myeloid training that protects male but not female mice from high fat diet induced disease. Here we demonstrate that ovarian derived hormones account for this sex specific difference. Ovariectomy of females prior to infection permits metabolic reprogramming of the myeloid lineage, with BMDM exhibiting carbon source flexibility for cellular respiration, and mice protected from systemic metabolic disease.

Vacuolization of hematopoietic precursors cells is a common future of several otherwise non-related clinical settings such as VEXAS, Chediak-Higashi syndrome and Danon disease. Although these disorders have a priori nothing to do with one other from a clinical point of view, all share abnormal vacuolization in different cell types including cells of the erythroid/myeloid lineage that is likely the consequence of moderate to drastic dysfunctions in the ubiquitin proteasome system and/or the endo-lysosomal pathway. Indeed, the genes affected in these three diseases UBA1, LYST or LAMP2 are known to be direct or indirect regulators of lysosome trafficking and function and/or of different modes of autophagy.

Background: The morbidity and mortality of sepsis remain high, and so far specific diagnostic and therapeutic means are lacking.

Objective: To screen novel biomarkers for sepsis.

Methods: Raw sepsis data were downloaded from the Chinese National Genebank (CNGBdb) and screened for differentially expressed RNAs.
