Arachidonic acid augments potassium currents in rat neocortical neurones.

We analysed the effects induced by arachidonic acid on voltage-dependent outward currents generated by rat neocortical neurones in culture in the presence of Na+ and Ca2+ channel blockers. These currents, recorded with whole-cell voltage-clamping techniques, were presumably carried by K+ and were characterized by an early transient and a late persistent component. Extracellular application of arachidonic acid (50 microM) enhanced both components of the voltage-dependent K+ response by 15-29% (n > 20 cells). These effects were reversible and not observed when a low dose (50 microM) of indomethacin was present in the bath (n = 7 cells). We conclude that one or more of the arachidonic acid metabolites of the cyclooxygenase pathway might be involved in the modulation of outward currents in neocortical cells in culture.

This study investigated the protective effects of the dietary polyphenol vanillic acid (VA) on dextran sulfate sodium-induced acute ulcerative colitis (UC) in mice, focusing on its impact on the gut microbiota and inflammatory responses. VA was supplemented following dextran sulfate sodium administration, and key indicators, including body weight, disease activity index, colon length, spleen index, and inflammatory markers, were assessed. VA supplementation significantly alleviated UC symptoms, preserved intestinal barrier integrity, and reduced pro-inflammatory cytokine levels.

Norvaline is a nonproteinogenic amino acid and an important food ingredient supplement for healthy food. In this study, dl-norvaline administration reduced body weight by more than 40% and improved glucose metabolism and energy metabolism in obese mice induced by a high-fat diet (HFD). Combination analysis of microbiome and metabolomics showed that dl-norvaline supplementation regulated gut bacteria structure, such as increasing beneficial bacteria (, , , , , , , and ) and decreasing harmful bacteria (, , , , , and ) and modulated the metabolites involved in arachidonic acid metabolism, thus further promoting short-chain fatty acid production and improving gut barrier, thereby inflammatory responses and oxidative stress were ameliorated.
