In the present study, we compared the ability of human neutrophils and monocytes to display oxygen-dependent cytotoxic responses at pH 7.4 and 6.2. Our results show that cytotoxicity induced by immune complexes (IC), zymosan, N-formyl-methionyl-leucyl-phenylalanine (FMLP) and concanavalin A (Con A) were markedly increased when they were carried out at pH 6.2 instead of pH 7.4. Cytotoxicity induced by phorbol myristate acetate (PMA), on the contrary, was significantly decreased at pH 6.2. It is noteworthy that cytotoxic responses induced by IC, zymosan and Con A were also increased when, 2 h after effector cell stimulation at pH 6.2, cytotoxicity was measured at pH 7.4. Finally, when we examined possible mechanisms involved in the augmentation of cytotoxicity, we observed that the oxidative response of IC-stimulated neutrophils, measured as chemiluminescence emission, was not increased at pH 6.2, on the contrary, it was significantly decreased. The relevance of these results is discussed.

Anti-tumor immunotherapy was rediscovered and rejuvenated in the last two decades with the discovery of CTLA-4, PD-1 and PD-L1 and the roles in inhibiting immune function and tumor evasion of anti-tumor immune response. Following the approval of the first checkpoint inhibitor ipilimumab against CTLA-4 in melanoma in 2011, there has been a rapid development of tumor immunotherapy. Furthermore, additional positive and negative molecules among the T-cell regulatory systems have been identified that that function to fine tune the stimulatory or inhibitory immune cells and modulate their functions (checkpoint modulators).

In this study, thiolated sodium alginate (SA) and hydrophilic, polymerizable Janus-type polyhedral oligomeric silsesquioxane (AS-POSS) are synthesized by introducing thiol and sulfonic acid groups, respectively. A series of pH-responsive SA/PEGDA/AS-POSS nanocomposite hydrogels are successfully prepared through Michael addition reactions between the thiol groups of thiolated sodium alginate and the double bonds in the molecular chains of AS-POSS and poly(ethylene glycol) diacrylate (PEGDA). This reaction proceeds rapidly under physiological conditions without requiring initiators or catalysts.

Novel circularly-polarized-luminescence (CPL) materials were designed and synthesized by integrating the intrinsic chirality of helicene with the acid/base-responsive properties of fluorescein. The synthesized helicene-fluorescein hybrids exhibit reversible switching between blue fluorescence under neutral conditions and red fluorescence under basic conditions. Furthermore, these hybrid compounds demonstrate unique chiroptical switching behavior with a rare base-triggered CPL sign inversion, where (P)-isomers show a negative CPL signal around 450 nm under neutral conditions and a positive signal around 620 nm under basic conditions, with three-fold enhanced dissymmetry factors (|glum|) under basic conditions.

The immunosuppressive microenvironment in pancreatic cancer, characterized by low tumor-specific T cells and excessive fibrosis, limits the effectiveness of immunotherapy. Here, three datasets and multi-immunofluorescence staining of tissue microarrays in pancreatic cancer indicate that mesothelin (MSLN) expression negatively correlates with cytotoxic T cells in tumor. Anti-MSLN antibody (αMSLN) treatment of pancreatic cancer in vivo can significantly increase T cell infiltration.
