Background: This in vitro study evaluated the efficacy of professional and home-use fluoride regimens for protecting irradiated enamel, undergoing pH cycling resembling xerostomia.

Methods: Sixty human premolar teeth were irradiated with a total dose of 70 Gy and subsequently sectioned into 3 × 3 cm enamel slabs. These slabs were randomly distributed into five groups (n = 12 per group): professional-use groups received fluoride varnish either weekly (FV1) or biweekly (FV2); home-use groups applied 5000 ppm (FT5) or 1450 ppm (FT) fluoride toothpaste; and a control group (control) received no treatment.

Single-cell genomics technologies have accelerated our understanding of cell-state heterogeneity in diverse contexts. Although single-cell RNA sequencing identifies rare populations that express specific marker transcript combinations, traditional flow sorting requires cell surface markers with high-fidelity antibodies, limiting our ability to interrogate these populations. In addition, many single-cell studies require the isolation of nuclei from tissue, eliminating the ability to enrich learned rare cell states based on extranuclear protein markers.

GABA receptors mediate prolonged inhibition in the brain and are important for keeping neuronal excitation and inhibition in a healthy balance. However, under excitotoxic/ischemic conditions, GABA receptors are downregulated by dysregulated endocytic trafficking and can no longer counteract the severely enhanced excitation, eventually triggering neuronal death.
