Effects of 10% O2 on the response of the phrenic neurogram to vagal (VS) and saphenous stimulation (SS) were studied in anesthetized and artificially ventilated newborn (group 1: 1-3 days old, n = 13; group 2: 8-14 days old, n = 14) and adult (group 3, n = 12) rabbits. In all animals during ventilation with 100% O2 or 10% O2 in N2, VS caused a reduction in the expiratory phase (Te). Te shortening in response to SS was unaffected by hypoxia in group 3, and was abolished in group 2. In group 1, only early hypoxia blocked the Te response to SS. The preserved Te response to VS on one hand, and abolished the effects of SS on the other, suggest that blockade of the somatic inputs is due to the age-dependent action of hypoxia in areas distant to the respiratory neurons.

Mechanisms contributing to the genesis of hypoglossal preinspiratory discharge.

Preinspiratory discharge manifests in the neuronal recordings of the pre-Bötzinger complex, parafacial respiratory group, retrotrapezoid nucleus, and Kölliker-Fuse nucleus, as well as the efferent neural discharge of respiratory-related nerves innervating upper airway musculature. This neural component of triphasic eupnea contemporaneously contributes to the genesis of native and originate respiratory rhythmic activity, as well as the preinspiratory component of efferent neural respiratory discharges. In the course of our investigations evaluating hypoglossal discharge in response to asphyxia, we noted a curious pattern of neural respiratory recovery following postasphyxia resuscitation in hypoglossal, vagal, and phrenic neurograms in unanesthetized decerebrate rats.

Glutamate is the predominant excitatory neurotransmitter in the ventral respiratory column; however, the contribution of glutamatergic excitation in the individual subregions to respiratory rhythm generation has not been fully delineated. In an adult, in vivo, decerebrate rabbit model during conditions of mild hyperoxic hypercapnia we blocked glutamatergic excitation using the receptor antagonists 2,3-dihydroxy-6-nitro-7-sulfamoyl-benzo[f]quinoxaline-2,3-dione (NBQX) and d(-)-2-amino-5-phosphonopentanoic acid (AP5). Disfacilitation of the preBötzinger Complex caused a decrease in inspiratory and expiratory duration as well as peak phrenic amplitude and ultimately apnea.

The role of the dorsolateral pons in the control of expiratory duration (Te) and breathing frequency is incompletely understood. A subregion of the pontine parabrachial-Kölliker-Fuse (PB-KF) complex of dogs was identified via microinjections, in which localized pharmacologically induced increases in neuronal activity produced increases in breathing rate while decreases in neuronal activity produced decreases in breathing rate. This subregion is also very sensitive to local and systemic opioids.
