NADPH diaphorase histochemistry was used to determine whether the rat anoocccygeus (AC) and retractor penis (RP) muscles are innervated by nerves capable of synthesizing nitric oxide. In both tissues, muscle fascicles were enclosed by a varicose plexus of NADPH diaphorase positive (ND+) fibers. Perikarya of neurons on the surface of the AC muscle were also intensely stained for NADPH diaphorase. Many AC-RP ganglion cells in the pelvic plexus, located by the retrograde tracer Fluorogold, also stained for the enzyme. However, a significant population of AC-RP neurons in this location remained unstained. These results provide further evidence that nitric oxide may be an important neurotransmitter in these tissues.

Nitric oxide synthases (NOSs) are heme-based monooxygenases that catalyze the NADPH-dependent oxidation of L-arginine to produce NO and L-citrulline. Over the past five years, the identification and characterization of NOS homologs in cyanobacteria have significantly advanced our understanding of these enzymes. Related to the inactive form of nitrile hydratase, NHase, that contains Fe(NO) within tripeptide NS binding environment, the NO transfer reactivity of (bis-mercaptoethane diazacycloheptane)Fe(NO) and (bis-mercaptoethane diazadimethylethane)Fe(NO) is compared to Co(NO) analogs. Acceptors of NO include cobalt octaethylporphyrin and the [(NS)M] dimeric precursors in the synthesis of the Fe(NO) and Co(NO) biomimetics. Qualitative rates are augmented by a definitive kinetic study finding that rates of NO transfer from (NS)M(NO) to [(NS)M'] are dependent on M and M' as well as the hydrocarbon N to N and N to S linkers.
