Human interferon alpha (HuIFN-alpha) preparations induced in human peripheral white blood cells by Sendai virus were analyzed for activity after ultrafiltration, ion exchange, metal ion chromatography and isoelectrofocusing. It was found that HuIFN-alpha forms complexes with various physical properties and activities. Some of the IFN-alpha copurify with high molecular weight complexes. However, the low molecular weight fraction represent bulk of IFN activity. In isoelectrofocusing experiments, the former corresponds to IFN-alpha which possesses high pI (pH regions 8.5-10.5). An additional form of IFN-alpha are not complexed and possess a pI of 5.5-7.0. Our nIFN-alpha preparation preserve many of natural IFN isospecies and may have higher therapeutic value than the recombinant IFN-alpha preparations.

Background & Aims: Eosinophilic esophagitis (EoE) is a chronic esophageal inflammatory disorder characterized by eosinophil-rich mucosal inflammation and tissue remodeling. Prior research has revealed the upregulation of interferon (IFN) response signature genes (ISGs) in biopsy tissue from EoE patients, but the specific cell types that contribute to this IFN response and the effect of interferons on the esophageal epithelium remain incompletely understood. Here, we use scRNA-seq to examine the expression of IFN and ISGs during EoE and explore how IFN-α and IFN-γ treatments affect epithelial function.

Background: Adding pembrolizumab, an anti-PD-1 antibody approved for treatment of head and neck squamous cell carcinoma (HNSCC) to neoadjuvant (induction-) chemotherapy utilizing docetaxel and cisplatin (TP) followed by radiotherapy may improve outcome in larynx organ-preservation (LOP) that is investigated in the European Larynx-Organ preservation Study (ELOS). As biomarkers for response to TP and pembrolizumab +TP are missing but may include cytokines, this work aims on determining cytokines potentially linked to outcome as prognostic markers sufficient to predict and/or monitor response to successful LOP.
Methods: Collagenase IV digests were generated from 47 histopathological confirmed HNSCC tumor samples and seeded in 96-well plates containing pembrolizumab, docetaxel, cisplatin either solely or in binary or ternary combination.
