A scanning electron microscope (SEM) survey assessed the quality of ultrastructural preservation of a wide range of tissues from the laboratory rat and from a limited number of botanical and fly-larval samples, air-dried from tetramethylsilane (TMS) after chemical fixation and dehydration. Comparisons were made with critical point drying (CPD) and air-drying from acetone. CPD produced good results in all cases. TMS gave acceptable results with 75%, and acetone with 31%, of rat tissues. Air-drying was unacceptable for the botanical and fly-larval samples. It is concluded that air-drying from TMS is generally useful for mammalian tissues in preparation for the SEM.

Purpose: This study aimed to evaluate the effectiveness of single versus group culture strategies for cumulus-oocyte complexes (COCs) derived from early antral follicles (EAFs), with the goal of optimizing culture conditions to increase oocyte availability for assisted reproductive technologies.
Methods: COCs isolated from EAFs (350-450 µm) from sheep ovaries were cultured in TCM199 medium supplemented with 0.15 µg/mL Zn as zinc sulfate, 10 IU/mL FSH, 10 ng/mL estradiol, 50 ng/mL testosterone, 50 ng/mL progesterone, and 5 µM Cilostamide.

The color and aroma of nectarines experience adverse effects from cutting, resulting in the fast senescence of fruit tissue. Therefore, 1-methylcyclopropene (1-MCP) was used to treat postharvest nectarines before cutting, and its effect on the surface browning and aroma alteration were investigated. The results indicated that 1-MCP restrained the soluble quinone (SQC) accumulation in fresh-cut nectarines by regulating the peroxidase (POD) and polyphenol oxidase (PPO) activities and the metabolism of phenolic compounds.

Cryopreservation, the preservation of tissues at subzero temperatures, is a mainstay of brain banking that allows for the storage of brain tissue without the use of chemical fixatives. This is particularly important for molecular studies that are incompatible with tissue fixation. However, brain tissue is vulnerable to various forms of damage during the cryopreservation process, in particular due to the phase transition of water from a liquid to a solid state with the formation of ice crystals, which can disrupt cellular morphology.
