Transforming growth factor-beta up-regulates human elastin promoter activity in transgenic mice.

We have recently developed transgenic mice which express approximately 5.2 kb of the human elastin promoter linked to the chloramphenicol acetyl transferase (CAT) reporter gene (J. Biol. Chem. 269:18072-18075, 1994). Previously, transforming growth factor-beta (TGF-beta) has been shown to enhance elastin gene expression, as determined at the mRNA and protein levels. To examine whether this enhancement could be explained by upregulation of the elastin promoter, TGF-beta 1 (100 ng) was injected subcutaneously into the transgenic animals. CAT activity in the skin of treated animals was elevated in a time-dependent manner up to approximately 10-fold after a single injection. These results suggest that the 5.2-kb up-stream segment of the human elastin gene contains cis-elements responsive to TGF-beta 1 in vivo.

Recent studies have attempted to characterize the layer-specific mechanical and microstructural properties of the aortic tissues in either normal or pathological state to understand its structural-mechanical property relationships. Although this cellular family is constituted by several distinct subtypes, dermal fibroblasts are crucial thanks to their ability to secrete pro-regenerative growth factors, extracellular matrix (ECM) proteins and their immune and anti-inflammatory role. Sophorolipids (SL), sophorosides (SS) and glucolipids (G), mono-unsaturated (C18:1) or saturated (C18:0), glycolipids derived from microbial fermentation of wild type or engineered yeast , constitute a novel sustainable class of bio-based chemicals with interesting physicochemical characteristics, which allow them to form soft diverse structures from hydrogels to vesicles, micelles or complex coacervates with potential interest in skin regeneration applications.

Thoracoabdominal aortic aneurysms (TAAAs) are rare but serious conditions characterized by dilation of the aorta characterized by remodeling of the vessel wall, with changes in the elastin and collagen content. Individuals with Marfan syndrome have a genetic predisposition for elastic fiber fragmentation and elastin degradation and are prone to early aneurysm formation and progression. Our objective was to analyze the medial collagen characteristics through histological, polarized light microscopy, and electron microscopy methods across the thoracic and abdominal aorta in twenty-five patients undergoing open surgical repair, including nine with Marfan syndrome.
