E12 and E47 are two helix-loop-helix transcription factors that arise by alternative splicing of the E2A gene. Both have been implicated in the regulation of immunoglobulin gene expression. We have now generated E2A (-/-) mice by gene targeting. E2A-null mutant mice fail to generate mature B cells. The arrest of B cell development occurs at an early stage, since no immunoglobulin DJ rearrangements can be detected in homozygous mutant mice. While immunoglobulin germline I mu RAG-1, mb-1, CD19, and lambda 5 transcripts are dramatically reduced in fetal livers of E2A (-/-) mice, B29 and mu degrees transcripts are present, but at lower levels. In addition, we show that Pax-5 transcripts are significantly reduced in fetal livers of E2A (-/-) mice. These data suggest a crucial role for E2A products as central regulators in early B cell differentiation.

Purpose To explore the diffusion characteristics of hypertrophic cardiomyopathy (HCM) using in vivo cardiac diffusion-tensor imaging (cDTI) and to determine whether cDTI could help identify abnormal myocardium beyond cardiac MRI findings of fibrosis and hypertrophy. Materials and Methods In this prospective study conducted from April to August 2023, participants with HCM and healthy volunteers were enrolled for cardiac MRI evaluation, including cine, late gadolinium enhancement (LGE), T1 mapping, and cDT imaging, using a 3.0-T scanner.

The replication-defective adeno-associated virus (AAV) is extensively utilized as a research tool or vector for gene therapy. The production process of AAV remains intricate, expensive, and mechanistically underexplored. With the aim of enhancing AAV manufacturing efficiencies in mammalian cells, we revisited the questions and optimization surrounding the requirement of the various adenoviral helper genes in enabling AAV production.

Cardiac diffusion tensor imaging (cDTI) is highly prone to image corruption, yet robust-fitting methods are rarely used. Single voxel outlier detection (SVOD) can overlook corruptions that are visually obvious, perhaps causing reluctance to replace whole-image shot-rejection (SR) despite its own deficiencies. SVOD's deficiencies may be relatively unimportant: corrupted signals that are not statistical outliers may not be detrimental.

We have introduced the floxed allele of Smoothened (Smo) carried by the mouse line Smo into the C57BL/6J strain by serial backcross. Recapitulation of the Smo null phenotype was confirmed by deleting the allele using E2a-cre and intercrossing heterozygous Smo ± mice. No homozygous mutant embryos were identified at E9.

This paper presents a non-linear finite element model (FEM) to predict the load-carrying capacity of three different configurations of elliptical concrete-filled steel tubular (CFST) short columns: double steel tubes with sandwich concrete (CFDST), double steel tubes with sandwich concrete and concrete inside the inner steel tube, and a single outer steel tube with sandwich concrete. Then, a parametric and analytical study was performed to explore the influence of geometric and material parameters on the load-carrying capacity of elliptical CFST short columns. Furthermore, the current study investigates the effectiveness of machine learning (ML) techniques in predicting the load-carrying capacity of elliptical CFST short columns.
