The opportunity of enhancing the sequencing efficiency by applying continuous stacking hybridization is considered. The approach is based on the increase of duplex length by continuous stacking hybridization of oligonucleotides added to solution (l-oligonucleotides) to oligonucleotides immobilized on matrix (L-oligonucleotides). An analysis of reconstruction efficiency for sequenced fragments up to length of 30000 nucleotides was made. Various combinations of L- and l-oligonucleotide length were considered. The results obtained enable one to evaluate the potentialities of the proposed method for various nucleotide matrices and the complexity of experiment. Use of continuous stacking hybridization permits a considerable increase of the length of sequenced DNA fragments. We offer the approach for resolving ambiguities in branching points, which occur because of long repeats. It is based on continuous stacking hybridization of several l-oligonucleotides which form a "chain" stabilized by mutual stacking interaction.

Pyrite is considered as an effective and environmentally friendly substrate in constructed wetlands (CW) for wastewater treatment, but its application in recirculation stacking hybrid constructed wetlands (RSHCW) has been scarcely studied. This study uses varying amounts of pyrite as the substrate in RSHCW, leveraging the recirculation of wastewater to alter microenvironments such as dissolved oxygen (DO) and pH, to explore the potential mechanisms of nitrogen (N) and phosphorus (P) removal in pyrite-based RSHCW. The results show that as the proportion of pyrite increases, the removal rate of total phosphorus (TP) in the effluent also increases (25%→58%), significantly enhancing the deposition of iron-bound phosphorus (Fe-P) on the substrate, thereby turning CW into a P reservoir.

In ambient mass spectrometry, the performance in direct in situ analysis applications has been hindered by the lack of efficient ion-transferring technique between the atmosphere pressure ionization source and the mass analyzer. Building upon the hybrid concept of a stack ring ion guide and multipole ion guide, this study proposes the concept of a reconfigurable twisted dipole ion guide (TDIG) that enables flexible ion transfer between atmosphere and vacuum. Initially, theoretical and numerical studies were conducted to understand the basic ion confining principle of the twisted dipole ion guide, revealing its unique merits in long-distance flexible ion transmission.
