Discriminative stimulus properties of 7-OH-DPAT, a dopamine D3-selective receptor ligand.

7-Hydroxy-N,N-di-n-propyl-2-aminotetralin (7-OH-DPAT) is a high affinity and selective ligand at dopamine D3 receptors. Rats were trained to discriminate 7-OH-DPAT (0.05 mg/kg) from drug vehicle using a food-reinforced (FR 10) two-lever operant procedure. Six out of nine rats learned to discriminate 7-OH-DPAT, requiring a mean of 29 training sessions. After the training dose was increased to 0.10 mg/kg the remaining three rats reached discrimination performance criterion. The 7-OH-DPAT stimulus was dose-dependent. Doses of 0.01, 0.03, 0.056, and 0.1 mg/kg 7-OH-DPAT produced 0, 11, 78, and 100% drug-correct lever selection, respectively, with the ED50 value calculated to be 0.038 mg/kg. These results demonstrate that the D3-selective ligand 7-OH-DPAT can control differential operant responding in rats on the basis of its discriminative stimulus properties.

Method: Study 1 ( = 41) followed an iterative approach to develop two implicit association tasks measuring implicit attitude (positive versus negative valence) and motivation (wanting versus not wanting) regarding mood-improving behaviours, followed by an evaluation of the content validity for target stimuli representing these behaviours. In Study 2 ( = 230), the tasks were assessed for stimulus classification ease (accuracy and speed) and internal consistency.

The link between serotonergic modulation and depression is under debate; however, serotonin reuptake inhibitors (SRIs) are still the first-choice medicine in this condition. Disturbances in time perception are also reported in depression with one of the behavioral schedules used to study interval timing, differential-reinforcement-learning-of-low-rate, having been shown to have high predictive validity for an antidepressant effect. Here, we introduce an IntelliCage research protocol of an interval bisection task that allows more ecologically valid and less time-consuming rodent examination and provides an example of its use to confirm the previously reported acute effect of an SRI, clomipramine, on interval timing (increase in bisection point, D50).

Background: The Qnox index is a novel monitor to quantify intraoperative nociception based on an electroencephalographic algorithm. We evaluated the ability of the Qnox index to discriminate noxious from non-noxious stimuli, respond to stimuli, and discriminate different levels of analgesia in patients under propofol anaesthesia with neuromuscular block.
