Immobilization and ultrastructure of alveolar type II cells.

We studied the ultrastructural changes in the alveolar type II cells from the lung of immobilized male Wistar rats divided into 6 groups: control; immobilized for 1, 12, 24 and 30 hours; immobilized for 30 hours and readapted for 5 hours. We used the routine methods of fixation through perfusion and electron microscopy. The results indicated progressive structural changes in the alveolar type II cells, correlating with a prolonged time of akinesia. Characteristic was the swelling affecting first the mitochondria and then all other organelles and the plasmalemma. Hypertrophy and hyperplasia of the lamellar bodies were clearly seen. The relatively quick recovery suggested the accelerated mitosis of the alveolar type II cells.

The role of surfactant proteins A and D (SP-A and SP-D) in lung clearance and translocation to secondary organs of inhaled nanoparticles was investigated by exposing SP-A and SP-D knockout (AKO and DKO) and wild type (WT) mice nose-only for 3 hours to an aerosol of 20 nm gold nanoparticles (AuNPs). Animals were euthanised at 0-, 1-, 7- and 28-days post-exposure. Analysis by inductively coupled plasma mass spectrometry (ICP-MS) of the liver and kidneys showed that extrapulmonary translocation was below the limits of detection.

Exemplified by successful use in COVID-19 vaccination, delivery of modified mRNA encapsulated in lipid nanoparticles provides a framework for treating various genetic and acquired disorders. However, lipid nanoparticles that can deliver mRNA into specific lung cell types have not yet been established.
