Chick central nervous system (CNS) expresses alpha-bungarotoxin (alpha Bgtx) receptors. We have recently reported the purification and characterization of two alpha Bgtx receptor subtypes, alpha 7 and alpha 7-alpha 8 from chick optic lobe (COL). In order to study whether other alpha Bgtx receptor subtypes are present in other areas of the chick CNS, as well as their developmental expression, we used anti-alpha 7 and anti-alpha 8 subunit-specific antibodies to study alpha Bgtx receptors at different developmental stages in COL, brain and retina. We found that only the alpha 7 and alpha 7-alpha 8 subtypes are present at all developmental stages in chick COL and brain, where they represent 90% of all the alpha Bgtx receptors at embryonic day 19 and 1 day post hatching (D1). In chick retina, an alpha 8 subtype representing 50% of all alpha Bgtx receptors at D1 is present in addition to the alpha 7 and alpha 7-alpha 8 subtypes, and the expression of this alpha 8 subtype increases during neurodevelopment.

Neuropathic pain is still a serious and unsolved health problem. The pathophysiological mechanisms underlying the constellation of symptoms that characterize COVID-19 are only incompletely understood. In an effort to fill these gaps, a "nicotinic hypothesis," which posits that nicotinic acetylcholine receptors (AChRs) act as additional severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) receptors, has recently been put forth. A key feature of the proposal (with potential clinical ramifications) is the suggested competition between the virus' spike protein and small-molecule cholinergic ligands for the receptor's orthosteric binding sites.

Alzheimer's disease (AD) is associated with chronic neurodegeneration often accompanied by elevated levels of the neurotoxic peptide amyloid-beta 1-42 (Aβ42) in the brain. Studies show that extracellular Aβ42 binds to various cell surface receptors including the human α7 nicotinic acetylcholine receptor (nAChR) and activates pathways of neurotoxicity leading to cell death. The α7 nAChR is thus considered a promising drug target for therapy against neurodegenerative disease such as AD.

Inflammatory pain is a chronic, persistent and serious disease that greatly impacts public health, which is often accompanied by allodynia, hyperalgesia, and spontaneous pain. It is evident that α7 nicotinic acetylcholine receptor (α7nAChR) plays a key role in cholinergic anti-inflammatory pathway and exhibits the inhibition of neuroinflammation in chronic pain. Trifluoro-icaritin (ICTF), a derivative of icaritin from the extract of a genus of Epimedium plant, is identified to possess profound anti-inflammatory activity.
