Amyloid fibril formation of α-synuclein (αSN) is a hallmark of synucleinopathies. Although the previous studies have provided numerous insights into the molecular basis of αSN amyloid formation, it remains unclear how αSN self-assembles into amyloid fibrils in vivo. Here, we show that αSN amyloid formation is accelerated in the presence of two macromolecular crowders, polyethylene glycol (PEG) (MW: ~10,000) and dextran (DEX) (MW: ~500,000), with a maximum at approximately 7% (w/v) PEG and 7% (w/v) DEX.

Introduction: Wound treatment is a significant health burden in any healthcare system, which requires proper management to minimize pain and prevent bacterial infections that can complicate the wound healing process.

Rationale: There is a need to develop innovative therapies to accelerate wound healing cost-effectively. Herein, two polymer-based nanofibrous systems were developed using poly-lactic-co-glycolic-acid (PLGA) and polyvinylpyrrolidone (PVP) loaded with a combination of an antibiotic (Fusidic acid, FA) and a local anesthetic (Lidocaine, LDC) via electrospinning technique for an expedited healing process by preventing bacterial infections while reducing the pain sensation.

Hydroxyapatite, renowned for its biocompatibility and osteoconductive properties, plays a fundamental role in bone regeneration owing to its resemblance to natural bone mineral, thus offering considerable potential for advancing tissue engineering strategies. In this article, the innovative integration of silicon ions into biogenic (bovine-derived) hydroxyapatite (SiBHA) via a tailored sol-gel process is reported. The resultant SiBHA scaffolds exhibited an interconnected microporous structure with a total porosity of 70% and pore dimensions ranging from 120 to 650 µm.
