Regulation of gene expression by transcription factors Ets-1 and Ets-2.

The ETS family of transcription factors have a DNA-binding domain in common that binds a core GGA(A/T) DNA sequence. A large number of proteins have now been identified that contain an ETS DNA-binding domain (see review by Wasylyk et al., 1993). Ets-1 was first described as the cellular homolog of v-ets. which is translated as a 135-kDa gag-myb-ets fusion protein from the replication-deficient retrovirus E26 in chickens. Ets-2 was subsequently described as a closely related protein that contains the highly conserved ETS DNA-binding domain. This paper considers the manner by which the two closely related genes, Ets-1 and Ets-2, apparently play distinct roles in embryo development and in the immune system of adult mice. Although both Ets-1 and Ets-2 transform fibroblasts (Seth et al., 1989), the temporal and tissue-specific expression patterns suggest that these two proteins play distinct biological roles and consequently transactivate different downstream cellular target genes.

Sialic acid-binding immunoglobulin-like lectin 15 (Siglec-15) has been identified as a crucial immune suppressor in human cancers, comparable to programmed cell death 1 ligand (PD-L1). However, the regulatory mechanisms underlying its transcriptional upregulation in human cancers remain largely unknown. Here, we show that the transcription factors ETS-1 and ETS-2 bound to the Siglec-15 promoter to enhance transcription and expression of Siglec-15 in hepatocellular carcinoma (HCC) cells and that transforming growth factor β-1 (TGF-β1) upregulated the expression of ETS-1 and ETS-2 and facilitated the binding of ETS-1 and ETS-2 to the Siglec-15 promoter.

A major challenge in the development of a gene therapy for hemophilia A (HA) is the selection of cell type- or tissue-specific promoters to ensure factor VIII (FVIII) expression without eliciting an immune response. As liver sinusoidal endothelial cells (LSECs) are the major FVIII source, understanding the transcriptional F8 regulation in these cells would help optimize the minimal F8 promoter (pF8) to efficiently drive FVIII expression. During gastrulation, Wnt and FGF signaling pathways cooperate to induce posterior structures. Wnt target expression around the blastopore falls into two main categories: a horseshoe shape with a dorsal gap, as in Wnt8 expression; or a ring, as in FGF8 expression. Using ChIP-seq, we show, surprisingly, that the FGF signaling mediator Ets2 binds near all Wnt target genes.
