Alcaligenes eutrophus utilizing nerolidol, a sesquiterpene alcohol, as the sole source of carbon contains an inducible NAD(P)(+)-linked secondary alcohol dehydrogenase (SADH). The enzyme was purified to homogeneity by a combination of salt precipitation, ion exchange and affinity matrix chromatographies. The apparent molecular mass of the enzyme was estimated to be 139 KDa with four identical subunits of 38.5 KDa. The enzyme carried out both oxidation and reduction reactions. At pH 5.5, enzyme catalyzed the stereospecific reduction of prochiral ketones to secondary alcohols. The pH optimum for the oxidation reaction was 9.5. NADP+ and NADPH were respectively preferred over NAD+ and NADH for oxidation and reduction reactions. Some of the properties of this enzyme were found to be significantly different from those thus far described.

Seventeen 3-alkylaminoquinoxaline-2(1)-thiones and 3-alkyloxyquinoxaline-2(1)-thiones were prepared by a novel thionation protocol from the readily available quinoxaline-2,3-dione in excellent overall yields. This protocol starts with the chlorination of dione using thionyl chloride to give 2,3-dichloroquinoxaline followed by the reaction with equimolar amounts of -nucleophiles (primary amines and secondary amines) or -nucleophiles (phenols and alcohols) to principally afford 2-alkanamino-3-chloroquinoxalines or 2-alkyloxy-3-chloroquinoxalines, respectively. The chloroquinoxalines reacted with the thionation reagent -cyclohexyl dithiocarbamate cyclohexyl ammonium salt in ethanol under reflux to principally give the corresponding quinoxalin-2-yl cyclohexylcarbamodithioate that finally rearranges to give the corresponding thiones in 76-93% overall yields.

An atmospheric oxygen-mediated oxidative coupling of primary and secondary alcohols for the synthesis of nitrogen-containing heterocycles has been developed. This method utilizes atmospheric oxygen as the sole, environmentally friendly oxidant to convert a variety of alkyl and aromatic primary alcohols into aldehyde equivalents, avoiding over-oxidation to carboxylic acids. Notably, these mild oxidation conditions are compatible with both primary and secondary alkyl alcohols as substrates.
