Purification of human and rat kidney aldose reductase.

In the present study we report on a rapid two-step affinity chromatographic procedure to purify aldose reductase from human and rat kidney papilla and inner medulla. This enzyme, which is responsible for sorbitol formation in the kidney, was purified 145-fold from rat and 76-fold from human kidneys by consecutive Blue Sepharose and Matrex Orange chromatography. SDS-PAGE showed a single band of 38 kD for the human enzyme and a doublet of similar molecular weight for the rat kidney aldose reductase. The enzyme was characterized by substrate specificity and kinetic constants found identical to that of other organs purified previously.

Sepsis is a systemic inflammatory response to infection, and its occurrence is associated with a poor prognosis in the context of multiorgan dysfunction syndrome (MODS). Although there are several animal models for the study of its etiology, the cecal ligation and puncture (CLP) model has been considered the "Gold standard" because it shows a high degree of similarity to the progression of human sepsis. Currently, it is one of the most frequently chosen options to search for therapeutic alternatives to diminish the progression and organ damage induced by sepsis.

Background: We investigated chitosan's protective effects against tertiary butylhydroquinone (TBHQ)-induced toxicity in adult male rats, focusing on cognitive functions and oxidative stress in the brain, liver, and kidneys.

Methods: Rats were divided into four groups (n = 8/group): (1) Control, (2) Chitosan only, (3) TBHQ only, and (4) Chitosan + TBHQ.

Results: TBHQ exposure led to significant cognitive impairments and increased oxidative stress, marked by elevated malondialdehyde (MDA) and decreased superoxide dismutase (SOD) and glutathione (GSH) levels.
