Clones of Langat TP-21 virus with neurovirulence for primates much lower than that of Elantsev 15-20/3 strain may be obtained by threshold dilutions method on mice. Neurovirulence for monkeys of L1-L5 clones of Langat TP-21 virus obtained at our laboratory was similar to neurovirulence of A.Sabin's type III vaccinal poliomyelitis virus. Attenuated clones of Langat virus are stable and retain protective properties in experiments with mouse protection from pathogenic viruses of tick-borne encephalitis complex.

Unlabelled: We devised a model system to study persistent infection by the tick-borne flavivirus Langat virus (LGTV) in 293T cells. Infection with a molecularly cloned LGTV strain produced an acute lytic crisis that left few surviving cells. The culture was repopulated by cells that were ~90% positive for LGTV E protein, thus initiating a persistent infection that was maintained for at least 35 weeks without additional lytic crises.

The human genome comprises of abundant DNA sequences related to endogenous retroviruses (ERV) and a variety of solitary long terminal repeats (LTRs). Substantial numbers of intact retroviral particles have been detected by electron microscopy in normal human placental villous tissue particularly in syncytiotrophoblast. Understanding the molecular structure, organisation and distribution of these ERV sequences may lead to elucidation of their possible dual function at the foetal-maternal interface; proliferation and differentiation of cytotrophoblast and induction of local pregnancy-associated immune suppression thus allowing survival of the foetal allograft.
