Background: The role of cytokines in trauma still has not been satisfactorily elucidated. Multiorgan failure (MOF) development should be also under the direction of cytokines, endotoxin, and other mediators.
Methods And Results: Therefore we prospectively studied 88 patients with multiple trauma admitted to Traumatological Hospital Brno from June 1, 1992 to May 31, 1993. Extent of the trauma was determined by Injury Severity Score (ISS), Revised Trauma Score (RTS), and TRISS methodology with probability of survival. In study patients was investigated concentrations of interleukin 1, 2, 6, and tumor necrosis factor (TNF). Of above mentioned cytokines were elevated only IL-6 levels at admission and significant correlation with ISS was found (r = 0.32; p < 0.01). MOF developed in 23 patients (12 of them died), but it was not possible to predict the MOF development nor surviving according to admission levels of cytokines. A significant difference was observed in IL-6 levels of MOF patients one day before death (439 +/- 111 ng/l) in comparison with MOF patients, who survived (132 +/- 88 ng/l, p < 0.001). None of 12 MOF patients with IL-6 concentrations above 400 ng/l survived.
Conclusions: We conclude that IL-6, less TNF, seems to play an important role in organism response to multiple trauma, and later elevation in these cytokines levels, especially IL-6 level, in MOF patients mean poor prognosis. We had found a significant correlation between initial IL-6 level and ISS. Other cytokines did not show changes during the study.

Background: Fulminant myocarditis (FM) is a potentially lethal disease with a wide spectrum of clinical presentation, thus making the diagnosis hard to depict. In cases where acute circulatory failure occurs venoarterial (VA) extracorporeal membrane oxygenation (ECMO) support is a valid management strategy, especially in the pediatric and adult patients. This study aims to report the results of VA ECMO for FM in our Institution.

Background: Inadequate treatment responses, chemotherapy resistance, significant heterogeneity, and lengthy treatment durations create an urgent need for new pancreatic cancer therapies. This study aims to investigate the effectiveness of gemcitabine-loaded nanoparticles enclosed in an organo-metallic framework under ketogenic conditions in inhibiting the growth of MIA-PaCa-2 cells.
Methods: Gemcitabine was encapsulated in Metal-organic frameworks (MOFs) and its morphology and size distribution were examined using transmission electron microscopy (TEM) and Dynamic light scattering (DLS) with further characterization including FTIR analysis.

Conventional solid/liquid electrochemical interfaces typically encounter challenges with impeded mass transport for poor electrochemical quantification due to the intricate pathways of reactants from the bulk solution. To address this issue, this work reports an innovative approach integrating a target-activated DNA framework nanomachine with electrochemically driven metal-organic framework (MOF) conversion for self-sacrificial biosensing. The presence of the target biomarker serotonin initiates the DNA framework nanomachine by an entropy-driven circuit to form a cross-linked nanostructure and subsequently release the Fe-MOF probe.

This study reports the development of an innovative electrochemical sensor based on organometallic framework nanostructures for detecting valganciclovir (VLCV). VLCV is employed in the treatment of cytomegalovirus retinitis in AIDS patients. Rational design of nanoarchitectures for electroactive materials is a crucial approach for boosting their electrocatalytic performance.

Disulfiram (DSF), primarily applied in the therapy for alcohol addiction, has been demonstrated to possess the promising capability of anti-tumor in many human cancers, including esophageal squamous cell carcinoma (ESCC). To date, almost all studies about DSF in ESCC are focusing on investigating either drug combinations or nanoparticle-based delivery systems. However, the exact molecular mechanisms mediating the response to DSF in ESCC are totally unknown.
