Therapeutic use of phospholipids in thrombocytopenic or thrombocytopathic patients.

Eight thrombocytopenic/pathic patients received an intravenous infusion of phospholipids. In vitro bleeding test (Thrombostat 4000), thrombelastography, and resonance thrombography were performed in order to show the hemostatic effect. In none of the cases did phospholipids exert an effect comparable to platelet transfusion. Two patients, brother and sister, had a severe anaphylactoid reaction after the administration of phospholipids.

Phospholipids are subject to continuous transformation, including enzymatic hydrolysis (with the participation of phospholipases A, C, and D) to free polyunsaturated fatty acids (PUFAs), which under the influence of cyclooxygenases (COX1/2), lipoxygenases (LOXs), and cytochrome P450 (CYPs P450) are metabolized to various classes of oxylipins, depending on the type of PUFA being metabolized and the enzyme acting.

Oxidative stress plays a critical role in postmenopausal osteoporosis, yet its impact on osteoblasts remains underexplored, limiting therapeutic advances. Our study identifies phospholipid peroxidation in osteoblasts as a key feature of postmenopausal osteoporosis. Estrogen regulates the transcription of glutathione peroxidase 4 (GPX4), an enzyme crucial for reducing phospholipid peroxides in osteoblasts.

The externalization of Phosphatidylserine (PS) from the inner surface of the plasma membrane to the outer surface of the plasma membrane is an emblematic event during apoptosis and serves as a potent "eat-me" signal for the efferocytosis of apoptotic cells. Although less well understood, PS is also externalized on live cells in the tumor microenvironment and on live virus-infected cells whereby it serves as an immune modulatory signal that drives tolerance and immune escape. Given the importance of PS in cancer immunology and immune escape, PS-targeting monoclonal antibodies have been characterized with promising immunotherapeutic potential.

A standardized polyphenol-enriched fraction (IPHRFPPEF) was formulated into a phospholipid complex (IPHRFPPEF-PC) to enhance oral bioavailability and evaluate stability, toxicity, and in vivo anti-inflammatory activity in Sprague Dawley rats. IPHRFPPEF was prepared from crude extract using XAD-HP7/Diaion-HP20 resin column chromatography and analyzed via HPLC and NMR. Total phenolic and flavonoid contents were quantified, with IPHRFPPEF showing higher values than the crude fraction.
