Overexpression of members of the type 1 receptor tyrosine kinase (c-erbB) family has been documented in many types of cancer. In the case of c-erbB1 (epidermal growth factor receptor) and c-erbB2, this has been closely linked with poor prognosis, and in particular is apparently associated with an invasive/metastatic phenotype and relative insensitivity to conventional therapies. The cell surface location of these molecules renders them attractive targets for a variety of immunotherapeutic strategies, some of which are showing promise in preclinical and early clinical trials.

Well characterized, stable, p16-defective canine mammary cancer (CMT) cell lines and normal canine mammary epithelial cells were used to investigate expression of the major breast cancer-specific hormone receptors estrogen receptor alpha (ER1) and progesterone receptor (PR) as well as luminal epithelial-specific proto-oncogenes encoding c-B-1 (epidermal growth factor receptor/EGFr), c-B-2/HER2, c-B-3, and c-B-4 receptors. The investigation developed and validated quantitative reverse transcriptase polymerase chain reaction assays for each transcript to provide rapid assessment of breast cancer phenotypes for canine cancers, based on ER1, PR, and c-B-2/HER2 expressions, similar to those in human disease. Roles for relatively underexplored c-B-3 and c-B-4 receptor expressions in each of these breast cancer phenotypes were also evaluated.

The protein kinase c-erbB-2 belongs to the family of receptor tyrosine kinase and is involved in oncogenesis. The present study predicts different phosphorylation sites of HER2/c-erbB-2 which are important in preventing or developing cancer, especially breast cancer. Sequence homology showed highest homology (77%) with epidermal growth factor receptor kinase domain.

δ-Catenin is a member of the p120 catenin family. Similar to p120ctn, δ-catenin contains nine central Armadillo repeats and binds to the juxtamembrane domain (JMD) of E-cadherin. In this study, we have investigated the immunohistochemical expression of endoglin (CD105), vascular endothelial growth factor (VEGF), epidermal growth factor receptor (EGFR-1) and epidermal growth factor receptor 2 (c-erbB-2) and their prognostic correlation in 13 cases of cervical adenocarcinomas with mucinous, endometrioid and serous type differentiation. Our study revealed that for uterine cervix adenocarcinoma the most intense angiogenic activity occurs at the invasion front of these tumors. In addition, we noticed a trend towards increased CD105 MVD values in those cases in which were recorded the highest VEGF and c-erbB-2 reactivity.
