The promoter of vertebrate U6 small nuclear RNA genes consists of a TATA box and a snRNA proximal sequence element (PSE), and the combination of these two elements directs RNA polymerase III transcription. We detected RNA polymerase II transcription as well as pol III transcription from the human U6 promoter in a HeLa nuclear extract. The pol II-specific transcription was independent of the PSE and dependent on the presence of the TATA box. Both pol III- and pol II-specific transcription were stimulated by addition of recombinant TATA-binding protein (TBP). We conclude that both pol III and pol II preinitiation complexes can assemble on the U6 promoter in vitro and could compete during the bona fide process in the cell.

Two strains of , identified based on morphology and phylogenetic analysis, were isolated from rocky desertification soils in Yunnan province. Phylogenetic analyses inferred from three loci (the internal transcribed spacer of the nuclear ribosomal RNA gene, β-tubulin and RNA polymerase II second-largest subunit) showed that the two strains formed a single clade and were introduced as a new species of , is characterized by having ampulliform or broadly fusiform conidiogenous cells and dark olivaceous-green, oblong-ellipsoidal conidia. Phylogenetically, is most closely related to , but it distinguishes the latter by longer and narrower conidia.

PIWI-interacting RNAs (piRNAs) are a class of small noncoding RNAs associated with PIWI proteins within the male germline, and they play significant roles in maintaining genome stability via the modulation of gene expression. The piRNAs are implicated in the progression of various cancers, but the simultaneous monitoring of multiple piRNAs remains a challenge. Herein, we construct a single-molecule biosensor based on polymerization-transcription-mediated target regeneration for the simultaneous one-pot detection of multiple piRNAs.

This mini-review examines the strategy of combining viral protein sequence conservation with drug-binding potential to identify novel antiviral targets, focusing on internal proteins of influenza A and other RNA viruses. The importance of combating viral genetic variability and reducing the likelihood of resistance development is emphasised in the context of sequence redundancy in viral datasets. It covers recent structural and functional updates, as well as drug targeting efforts for three internal influenza A viral proteins: Basic Polymerase 2, Nuclear Export Protein, and Nucleoprotein.
