Does solute stereochemistry influence percutaneous penetration?

The stratum corneum, the rate-limiting barrier to percutaneous penetration, is made up of several components, principally keratin and ceramides. These are potential sources of chiral discrimination that could result in differential diffusion rates, dependent upon the stereochemistry of the solute. Although binding to keratin can occur it is not a stereoselective process [percent binding to solubilised epidermal keratin: (R)-propranolol 7.9 +/- 1.7, (S)-propranolol 8.3 +/- 2.0]. On the other hand, studies with ceramide monolayers produced qualitative evidence of dose-dependent stereoselective interaction when the pure diastereomers of ephedrine were present in the aqueous subphase which suggested that differences in diffusion rates might occur in skin. However, the differences in permeation rates in vitro for these diastereomers through human skin were not statistically different [(+)-(1S,2R)-ephedrine 119.1 +/- 2.6 micrograms/cm2, (-)-(1R,2S)-ephedrine 107.0 +/- 3.9 micrograms/cm2, 12 h]. Time averaging, involving contributions from binding to all lipid headgroups present in the intercellular channels, may obscure specific differential interactions. Further, any stereospecific interaction may be subtle and readily overwhelmed if diffusant concentration is greater than the capacity of the skin to differentiate between stereoisomers. Evidence for intrinsic stereoselectivity in skin permeation has therefore yet to be obtained.

Objectives: The Scrotal and Penile Imaging Working Group (SPIWG) of the European Society of Urogenital Radiology (ESUR) aimed to formulate recommendations on the imaging modalities and minimal technical requirements for abdominopelvic imaging in the follow-up of adult patients treated for testicular germ-cell tumors (TGCT).

Methods: The SPIWG members performed an extensive literature search, reviewed the current clinical practice, and reached a consensus based on the opinions of experts in the field.

: A previous study investigated the in vitro release of methylene blue (MB), a widely used cationic dye in biomedical applications, from nanocellulose/nanoporous silicon (NC/nPSi) composites under conditions simulating body fluids. The results showed that MB release rates varied significantly with the nPSi concentration in the composite, highlighting its potential for controlled drug delivery. To further analyze the relationship between diffusion dynamics and the MB concentration, this study developed a finite element (FE) method to solve Fick's equations governing the drug delivery system.

Scanning electron microscopy (SEM) revealed a complex network of interconnected pores within the composite materials, with varying pore sizes contributing to the material's overall properties. Acoustic characterization was conducted using a two-microphone impedance tube.

This paper summarizes the main findings of a study which aimed to examine the electrochemical oxidation of homovanillic acid (HVA), the final metabolite of dopamine. A pencil graphite electrode (PGE) was used as working electrode and the measurements were performed by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The type and the composition of the graphite leads used as PGE, the pH of the supporting electrolyte, as well as the scan rates were optimized by CV.
