Monoclonal antibodies specifically recognizing rat transferrin were produced. Optimal conditions of fixation and carrying out immunocytochemical reaction were determined. A combined method is proposed based on the indirect immunoperoxidase reaction and Feulgen procedure to be used for studying transferrin contents in hepatocytes with different ploidy. The transferrin content increased proportionally to the cell ploidy. It is concluded that the cytophotometric method may be used for a quantitative study of transferrin content in isolated rat hepatocytes.
