An attempt has been made to reveal simultaneously both mast and ECL cells in the fundic mucosa of some laboratory animals and man. In Bouin fluid fixed specimens, toluidine blue pH 5.0 and alcian blue pH 1.0 failed to reveal mucosal mast cells in rats and mice only. In those animals mucosal mast cells became demonstrable in Carnoy fluid fixed tissues after staining with alcian blue pH 1.0. A double staining technique has been applied using Grimelius silver method followed by staining either with toluidine blue after acid hydrolysis or with alcian blue. Both mast and ECL cells became visible showing here and there their close arrangement. The latter might be a point for some functional relations between both cell types.

The purpose of this study was to evaluate EpiColon, a novel human organotypic 3D colon microtissue prototype, developed to assess colonic drug disposition, with a particular focus on permeability ranking, and compare its performance to Caco-2 monolayers. EpiColon was characterized for barrier function using transepithelial electrical resistance (TEER), morphology via histology and immunohistochemistry, and functionality through drug transport studies measuring apparent permeability (P). Cutoff thresholds for the permeability of FITC-dextran 4 kDa (FD4), FITC-dextran 10 kDa (FD10S), and [C]mannitol were established to monitor microtissue integrity.

One of the main endpoints for the evaluation of Developmental and Reproductive Toxicology (DART) studies is the determination of potential effects of a test substance on the skeleton during foetal development. In the course of a DART study according to the OECD 414 guideline, 400-500 gestational day 20-old (GD20), alizarin red and alcian blue-stained (ARAB) rat foetuses have to be assessed by a teratology expert, which is a time consuming and sub-optimally documented process. We have developed a method which allows for a standardised, comprehensive, quick and easy to perform, head-to-toe digital documentation of ARAB-stained GD20 rat foetuses.
