Blastomeres from eggs of the killifish, Fundulus, were mechanically dissociated and reaggregated by pelleting in a simple saline solution. Formation of gap and tight junctions was followed by electron microscopy of freeze-fracture replicas. Five to eight min after pelleting, neither new nor old junctions were observed. After 10-15 min, small gap junctions were found, but these were not associated with distinct formation plaques. Larger gap junctions were observed after 45 min, and the images were consistent with growth by accretion of intramembrane particles. In aggregates, after 20 min or more, tight junctions were much more commonly found than in intact blastulae, and it seemed likely that they were being formed by cells that were not doing so in the intact embryo. Initial stages consisted of short strands that appeared to grow in length. Also, more elaborate junctions were seen than occur in situ. Particle-free membrane often occurred near incomplete junctions, and large junctions like those in situ separated particle-rich from particle-free membrane. In this system, the formation of both gap and tight junctions occurs with shorter latency, and is more precisely timed, than heretofore described.

The epithelial barrier in different organs is the first line of defense against environmental insults and allergens, with type 2 immunity serving as a protective function. Genetic factors, and biological and chemical insults from the surrounding environment altered regulate epithelial homeostasis through disruption of epithelial tight junction proteins or dilated intercellular spaces. Recent studies suggest that epithelial barrier dysfunction contributes to pathologic alteration in diseases with type 2 immune dysregulation including (but not limited to) atopic dermatitis, prurigo nodularis, asthma, chronic rhinosinusitis with nasal polyps, and eosinophilic esophagitis.

Yak is a large plateau mammal with low reproduction rate, while cryptorchidism is a major reproductive disorder associated with infertility in highland yaks. To better understand the occurrence of cryptorchidism and its regulatory mechanisms in yaks, we conducted a multi-omic analysis, and screened a total of 4456 differentially expressed genes (DEGs) and 332 differentially expressed proteins (DEPs) between normal and unilateral cryptorchid testes of yaks using high-throughput transcriptome sequencing (RNA-seq) and Tandem Mass Tag (TMT)-based proteomics techniques, with testes from yaks with cryptorchidism as the target. Enrichment analysis revealed that the DEGs were associated with cell growth, sperm motility, immune regulation, and intercellular tight junctions, and were mainly enriched in pathways related to cell differentiation; amino acid, sugar, and lipid metabolism; cell adhesion, and hypoxia tolerance.

In the context of transdermal delivery, favoring the drug permeability of epithelia through convenient formulations would open new opportunities for local versus systemic drug delivery, envisaging higher patient comfort and an enhanced therapeutic effect. Ligands of tight junctions are interesting agents that enhance epithelial permeability by relaxing the protein complexes that form them. The C-terminal domain of enterotoxin (c-CPE), which binds claudins, one of the tight junction (TJ) components, has been explored here as a functional domain in modular recombinant proteins, to evaluate its ability to self-promote its paracellular epithelial passage in a Caco-2 cell monolayer model.
