CRISPR-Cas12a is widely used for genome editing and biomarker detection since it can create targeted double-stranded DNA breaks and promote non-specific DNA cleavage after identifying specific DNA. To mitigate the off-target DNA cleavage of Cas12a, we previously developed a Cas12a variant (FnoCas12a ) by introducing double proline substitutions (K969P/D970P) in a conserved helix called the bridge helix (BH). In this work, we used cryogenic electron microscopy (cryoEM) to understand the molecular mechanisms of BH- mediated activation of Cas12a.

Given the large volumes of sensitive information transmitted over the Internet, digital signatures are essential for verifying message authenticity and integrity. A key challenge is minimizing computationally intensive operations, such as modular inverses, without compromising security. In this research, we propose the DSADHπ algorithm, which introduces a confusion step directly into the signature itself, rather than only applying it to the message, using a dynamic substitution box.

Viologen derivatives feature two reversible one-electron redox processes and have been extensively utilized in aqueous organic flow batteries (AOFBs). However, the early variant, methyl viologen (MVi), exhibits low stability in aqueous electrolytes, restricting its practical implementation in AOFB technology. In this context, leveraging the tunability of organic molecules, various substituents have been incorporated into the viologen core to achieve better stability, lower redox potential, and improved solubility.

Dung Beetle algorithm is an intelligent optimization algorithm with advantages in exploitation ability.
