Complexes of carminomycin with DNA were prepared from lyophilized carminomycin and high polymer DNA of sturgeon milt. The complex formation was evident from a batochromic shift in the maxima at 450-600 nm and an increase in the absorption level at 545 nm. The carminomycin complexes with DNA had a relatively low general toxicity and higher antileukemic activity in studies on mice with tumor L-1210 as compared to the use of carminomycin alone. The maximum increase in the average life time of the animals was observed when a complex with a molar ratio of carminomycin and DNA of 1:10 (with respect to nucleotides) was used. It was found that the complex of carminomycin and DNA with a ratio of the components of 1:20 had a more pronounced effect on the animal hemopoiesis as compared to the use of carminomycin alone. It resulted in a more significant decrease in the number of the bone marrow myelokariocytes and peripheral blood erythrocytes and leukocytes.

KRN 8602 (MX2) is a novel morpholino anthracycline derivative having the chemical structure 3'-deamino-3'-morpholino-13-deoxo-10-hydroxycarminomycin hydrochloride. To investigate the mechanisms of resistance to MX2, we established an MX2-resistant phenotype (K562/MX2) of the human myelogeneous leukaemia cell line (K562/P), by continuously exposing a suspension culture to increasing concentrations of MX2. K562/MX2 cells were more resistant to MX2 than the parent cells, and also showed cross-resistance to etoposide and doxorubicin.

New TFO conjugates containing a carminomycinone-derived chromophore.

Conjugates obtained by linking the anthracycline intercalating chromophore to triple helix forming oligonucleotides (TFOs) have been used in a physicochemical study of the stability of triple helices with DNA sequences of pharmacological relevance. The intercalating moiety is represented by carminomycinone derivatives obtained upon O-demethylation and hydrolysis of the glycosidic linkage of daunomycin followed by the introduction of an alkylating residue at two different positions. Results of experiments with a polypurinic region present in the multidrug resistance (MDR) gene indicate that the stability of the triple helix is significantly enhanced by replacement of C's with (5-Me)C's in the TFO sequences tested.

Structural requirements for the formation of anthracycline-DNA adducts.

A series of anthracyclines (comprising carminomycins I, II and III, and barminomycin) were tested for their ability to react with DNA to form site-specific adducts using an in vitro transcription assay. The requirement for drug activation by formaldehyde was also assessed using a transcription assay and HPLC analysis of GC-containing oligonucleotide duplexes. In the absence of formaldehyde, barminomycin was the most reactive compound and carminomycin I the least reactive.
