Spatially resolved transcriptomics has made it possible to study the subcellular organization of mRNA, a critical aspect of cellular function. However, there is a dearth of analytical tools to identify and interpret the functional significance of subcellular spatial distribution patterns. To address this, we present CellSP, a computational framework for identifying, visualizing, and characterizing consistent subcellular spatial patterns of mRNA.

Antibody ligation of HLA class II induces YAP nuclear localization and formation of cytoplasmic YAP condensates in human endothelial cells.

Immunohorizons

January 2025

Vatche and Tamar Manoukian Division of Digestive Diseases, Department of Medicine, David Geffen School of Medicine, University of California, Los Angeles, CA, United States.

Antibody (Ab) crosslinking of HLA class II (HLA II) molecules on the surface of endothelial cells (ECs) triggers proliferative and prosurvival intracellular signaling, which are implicated in promoting chronic Ab-mediated rejection (cAMR). Despite the importance of cAMR in transplant medicine, the mechanisms involved remain incompletely understood. Here, we examined the regulation of yes-associated protein (YAP) nuclear cytoplasmic localization and phosphorylation in human ECs challenged with Abs that bind HLA II, which are strongly associated with cAMR.

Integrative study of subcellular distribution, chemical forms, and physiological responses for understanding cadmium tolerance in two garden shrubs.

J Plant Physiol

January 2025

Beijing Forestry University, National Engineering Research Center for Floriculture, Beijing Laboratory of Urban and Rural Ecological Environment, Beijing, 100083, China. Electronic address:

Urban ornamental shrubs have significant potential for restoring cadmium (Cd)-contaminated soil. The Cd enrichment characteristics and tolerance mechanisms of Buxus sinica and Ligustrum × vicaryi were investigated through a simulated pot pollution experiment. Specifically, the Cd content and accumulation in different plant tissues, the subcellular distribution and chemical forms of Cd in the roots, and the effects of Cd on the ultrastructure of root cells under various Cd concentrations (0, 25, 50, 100, and 200 mg kg⁻) were analyzed.

Genome-wide identification of the Sec14 gene family and the response to salt and drought stress in soybean (Glycine max).

BMC Genomics

January 2025

Henan Collaborative Innovation Center of Modern Biological Breeding, College of Agronomy, Henan Institute of Science and Technology, Xinxiang, 453003, China.

Background: The Sec14 domain is an ancient lipid-binding domain that evolved from yeast Sec14p and performs complex lipid-mediated regulatory functions in subcellular organelles and intracellular traffic. The Sec14 family is characterized by a highly conserved Sec14 domain, and is ubiquitously expressed in all eukaryotic cells and has diverse functions. However, the number and characteristics of Sec14 homologous genes in soybean, as well as their potential roles, remain understudied.

Exposure to environmentally relevant levels of DEHP during development modifies the distribution and expression patterns of androgen receptors in the anterior pituitary in a sex-specific manner.

Chemosphere

January 2025

Consejo Nacional de Investigaciones Científicas y Tecnológicas (CONICET), Instituto de Investigaciones en Ciencias de La Salud (INICSA), Córdoba, Argentina; Universidad Nacional de Córdoba, Facultad de Ciencias Médicas, Centro de Microscopía Electrónica. Córdoba, Argentina. Electronic address:

DEHP is a prevalent phthalate with wide industrial applications and well-documented endocrine-disrupting effects, including the potential disruption of AR signaling in different tissues. The present study aimed to investigate the effects of gestational and lactational exposure to environmentally relevant DEHP concentrations on AR expression and subcellular localization in the pituitary gland, the master endocrine organ, with a focus on gonadotroph cells by in vivo and in vitro approaches. After DEHP exposure during gestation and lactation, a sex-specific modulation was detected in AR-positive pituitary cells and AR protein expression as assessed through flow cytometry and western blot.
