An improved method has been developed for determining internal insect infestation of wheat kernels. The method involves acid hydrolysis of cracked wheat, wet sieving to remove the acid, transferring to a 2 L Wildman trap flask, deaeration by boiling, and treatment with Tween 80-Na4EDTA. Insects are extracted with light mineral oil. Reports from 6 collaborators showed that recovery was 95.92% for adult insect heads and 97.22% for larvae by the proposed method as compared with 87.05% and 6.12%, respectively, by the official method. The method has been adopted official first action.

The Lepidopteran pest Trichoplusia ni and the parasitoid wasp Trichogramma brassicae represent a fascinating biological system, important for sustainable agricultural practices but challenging to observe. We present a nondestructive method based on micro-CT scanning technology (CT: computed tomography) for visualizing the internal parts of caterpillar embryos and of emerging parasitoids from infected eggs. Traditional methods of microscopic observation of the opaque egg contents require staining or dissection.

Aphids exhibit a unique reproductive strategy known as pseudoplacental viviparity, in which embryos develop internally and are thought to receive nutrients such as sugars and amino acids directly from the maternal hemolymph through an ovariole sheath, bypassing the need for traditional yolk storage. This system enables viviparous aphids to adapt to diverse and potentially stressful environments by transmitting maternal environmental cues that influence transgenerational plasticity. However, the mechanisms underlying this nutrient-mediated plasticity are poorly understood.
