Amino acidic composition, dispersion of optic rotation, differential temperature-perturbation spectra, molecular weight and terminal amino acids are studied for certain isoelectric fractions of human serum albumin obtained by means of isoelectric focusing in the borate-polyol system. The isolated three isoelectric fractions with pI 4.7, 4.9 and 5.1 revealed no significant differences in the composition of amino acids. They also did not differ in molecular weight and N- and C-terminal amino acids. However certain differences in the conformation and number of perturbed thyrosils are detected by means of spectral methods.

Timely identification of highly pathogenic bacteria is crucial for efficient mitigation of the connected harmful health effects. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) of intact cells enables fast identification of the microorganisms based on their mass spectrometry protein fingerprint profiles. However, the MALDI-TOF MS examination must be preceded by a time-demanding cultivation of the native bacteria to isolate representative cell samples to obtain indicative fingerprints.

Introduction: The identification of effective, selective biomarkers and therapeutics is dependent on truly deep, comprehensive analysis of proteomes at the proteoform level.

Methods: Bovine serum albumin (BSA) isolated by two different protocols, cold ethanol fractionation and heat shock fractionation, was resolved and identified using Integrative Top-down Proteomics, the tight coupling of two-dimensional gel electrophoresis (2DE) with liquid chromatography and tandem mass spectrometry (LC-MS/MS).

Results And Discussion: Numerous proteoforms were identified in both "purified" samples, across a broad range of isoelectric points and molecular weights.

This study aimed to assess pumpkin leaves as a protein source and determine the feasibility of these proteins to form complexes with alginate for the encapsulation of folic acid. Different isolation protocols, two based on isoelectric precipitation (one with thermal pretreatment and the other with alkali pre-extraction) and one based on stepwise precipitation with ammonium sulfate, were compared regarding the yield and structural properties of the obtained leaf protein concentrates (LPC). The highest purity of protein was achieved using the thermal-acid protocol and the salting-out protocol at 40% saturation.
