To increase the safety of stroma-free hemoglobin solution (SFH) as an artificial oxygen carrier source, we investigated the effect of heat treatment on virus inactivation in hemoglobin solution. The hemoglobin solution spiked with vesicular stomatitis virus (VSV) was treated at 60 degrees C for 1 hr under either an air or CO atmosphere. VSV was inactivated at >5.

Chemical characterization of pyridoxalated hemoglobin polyoxyethylene conjugate.

Biochim Biophys Acta

January 2000

Apex Bioscience Inc., P.O. Box 12847, Research Triangle Park, NC 27709, USA.

Pyridoxalated hemoglobin polyoxyethylene conjugate (PHP) was developed in the 1980s as an oxygen carrier and is now under development for treatment of nitric oxide-dependent, volume refractory shock. PHP is made by derivatizing human stroma-free hemoglobin with pyridoxal-5-phosphate and polyoxyethylene (POE). A unique aspect of using POE for modification is that unlike its mono-methoxy polyethylene glycol (PEG) relatives, POE is bifunctional.

The synthesis and hemoglobin cross-linking studies of a novel organic reagent, bis[2-(4-carboxyphenoxy)carbonylethyl]phosphinic acid (BCCEP; 2) has been reported. The reagent was synthesized in four steps from hydroxybenzoic acid. The tri-sodium salt of BCCEP was employed to cross-link oxyHb, and the product was purified by DEAE-cellulose chromatography.

Characteristics of Hb-vesicles and encapsulation procedure.

Artif Cells Blood Substit Immobil Biotechnol

January 1995

Department of Polymer Chemistry, Waseda University, Tokyo, Japan.

The performance of Hb-vesicles depends on the weight ratio of Hb to lipid ([Hb]/[Lipid]). This value is improved by lowering the number of bilayer membrane of the vesicle and raising the concentration of Hb in the interior of the vesicle. Maximum [Hb]/[Lipid] ratio was obtained at ca.

Application of aqueous two-phase systems in separation/purification of stroma free hemoglobin from animal blood.

Artif Cells Blood Substit Immobil Biotechnol

January 1995

Department of Chemical Engineering, National Tsing Hua University, Hsinchu, Taiwan, ROC.

Aqueous two-phase system (ATPS) is suitable for treatment of biological materials. It has been traditionally employed in the separation of product from fermentation broth, and in the extraction of large-molecule proteins from colloid solution. The separation method based on ATPS features simple, rapid and continuous operation with low equipment cost.
