Leber congenital amaurosis (LCA) is a sight-threatening inherited retinal disorder (IRD) caused by numerous genetic mutations. Multi-characteristic opsin (MCO)-based optogenetic therapy allows the recruitment of residual cells of the retina in LCA for alternative vision transduction while being mutation-agnostic. Using mice, we investigated the efficacy of an adeno-associated virus2 (AAV2)-transduced ambient light-activatable MCO (MCO-010) containing a metabotropic glutamate receptor-6 bipolar cell-specific promoter/enhancer.

Article Synopsis
  • The prion protein (PrP) is linked to neurodegenerative prion diseases, but its role in healthy cells, particularly in the retina, is unclear.
  • In this study, PrP was found at the junctions of photoreceptors and bipolar cells, indicating a possible involvement in synaptic function.
  • Deleting the PrP gene in mice led to significant changes in electrical responses (ERG) of photoreceptors and bipolar cells, especially in cone-dominant pathways, suggesting functional impacts without altering retinal structure.

Background: Rooms illuminated by "black light" (<400 nm wavelength) has become popular, but there is not enough scientific evidence to support its implementation. This study aims to assess the effects of violet light (392 nm) on the circadian rest-activity rhythm and the visual system through animal experimentation.

Materials And Results: Five groups of four mice were exposed to different white light, violet light, and dark periods, and their circadian rhythm was analyzed by measuring the circadian period using rest-activity cycles.

Purpose: To study the inner and outer retinal functions using a full-field electroretinogram (ERG) before and after intravenous chemotherapy (IVC) in children with retinoblastoma (RB).

Methods: Of the 11 eyes, seven had RB and four were normal. All children were examined under anesthesia using a handheld ERG machine with a standard protocol - light-adapted single-flash ERG (fERG), photopic single-flash 3.

The harmful effects of blue light on the retina and health issues attributed to flickering light have been researched extensively. However, reports on the effects of flickering blue light at a frequency in the visible range on the retina are limited. This study aimed to non-invasively investigate the structural and functional changes in mice retinas following exposure to flickering blue light.
