Objectives: To evaluate the protective effect of cystatin (r-Cystatin) in a mouse mode of "two-hit" sepsis.

Methods: Sixty male C57BL/6 mice randomized equally into sham-operated group, protein group, "two-hit" modeling group, and protein intervention group. In the former two groups, the mice received an intraperitoneal injection of 100 μL PBS followed by exposure of the cecum and then by intraperitoneal injection of 100 μL PBS or 25 μg r-Cystatin 30 min later; In the latter two groups, 100 μL PBS containing LPS (5 mg/kg) was injected intraperitoneally 24 h before cecal ligation and puncture (CLP), and 100 μL PBS or 25 μg r-Cystatin were injected 30 min after CLP.

Role of Arg1 ILC2s and ILCregs in gestational diabetes progression.

Sci Rep

Department of Obstetrics and Gynecology, Reproductive Medicine Center, The First Affiliated Hospital of Anhui Medical University, Wanshui Road No. 120, Hefei, 230000, Anhui, China.

Innate lymphoid cells (ILCs) are a newly discovered subset of immune cells that are responsible for regulation of the immune microenvironment. In particular, the ILC categories ILC2s and regulatory ILCs (ILCregs) are associated with immunosuppression and chronic inflammation. Chronic low-grade inflammation leads to insulin resistance, a major etiological factor in gestational diabetes mellitus (GDM).

Aims: Carboxylesterase (Ces)1f is implicated in protection against hepatic inflammation, but it is unclear whether the enzyme has an influence in polarization of Kupffer cells (KCs), the innate immune cells mediating hepatic inflammatory injury including acute liver failure (ALF). In the present study, we aim to explore KC polarization induced by Ces1f in mice with lipopolysaccharide/D-galactosamine (LPS/D-GalN)-induced ALF. We adopted a novel delivery system, β-1,3-D-glucan-encapsulated Endoporter-siRNA particles, to specifically target KC Ces1f knockdown via tail vein injection in mice.

The role of arginase estimation to predict hypertensive disorder of pregnancy at an earlier stage.

J Pak Med Assoc

October 2024

Department of Gynaecology and Obstetrics, Islamic International Medical College, Riphah International University, Islamabad, Pakistan.

Article Synopsis
  • The study aimed to analyze the role of serum arginase levels as an early predictor for pregnancy-related hypertension and preeclampsia in women between 20-25 weeks of gestation.
  • A total of 90 pregnant women were divided into three groups based on their health status: a control group (no complications), a group with pregnancy-induced hypertension, and a group with preeclampsia, with various serum markers measured for each group.
  • Results showed significant differences in serum arginase and other markers among the groups, indicating that elevated serum arginase levels could potentially help identify women at risk for hypertensive disorders during pregnancy.

Maresin 1 alleviates neuroinflammation and cognitive decline in a mouse model of cecal ligation and puncture.

Department of Cardiology, Xiangya Hospital, Central South University, Changsha 410008.

Objectives: Inflammation in the central nervous system plays a crucial role in the occurrence and development of sepsis-associated encephalopathy. This study aims to explore the effects of maresin 1 (MaR1), an anti-inflammatory and pro-resolving lipid mediator, on sepsis-induced neuroinflammation and cognitive impairment.

Methods: Mice were randomly assigned to 4 groups: A sham group (sham operation+vehicle), a cecal ligation and puncture (CLP) group (CLP operation+vehicle), a MaR1-LD group (CLP operation+1 ng MaR1), and a MaR1-HD group (CLP operation+10 ng MaR1).
