The results of the isolation of antigenic fractions and their complexes from Cl. oedematiens toxin and the study of their antigenic properties are presented. The purified, concentrated toxin has been found to consist of high- and low-molecular antigens differing in immunogenicity and affinity to aluminium hydroxide used as sorbent. Preparations obtained from low-molecular antigens easily adsorbed by the low doses of the sorbent possessed high specific and immunogenic activity.

The reactor technology for obtaining suspensions of Staphylococcus aureus (strain Cowan 1) with active protein A was developed. On the basis of the sensitization of bacterial suspensions with antitoxic rabbit sera staphylococcal reagents to diphtherial, gas gangrene (perfringens, oedematiens), botulinic toxins-toxoids were obtained and for the first time the coagglutination test with toxoids was carried out. More sensitive "specifically directed" reagents for the coagglutination test were obtained; for this purpose bacterial suspensions were sensitized with pure antibodies isolated from rabbit immune sera by immunosorption.

The alpha-toxin of Clostridium oedematiens type A was purified from culture filtrate by two steps of column chromatography and repeated gel filtration. The purified alpha-toxin proved homogeneous in polyacrylamide gel electrophoresis and agar gel double diffusion. The molecular weight of the alpha-toxin was estimated at 280,000 by sodium dodecyl sulfate polyacrylamide gel electrophoresis and at 260,000 by gel filtration on a Sephadex G-200 column.

To re-evaluate the current potency tests of C. oedematiens anti-toxin that titrates only the anti-lethal activity in mice, investigations were made on neutralization of the lethal, edematizing, and hemolytic activities with different preparations of the antitoxin and on their therapeutic effects. The anti-lethal and anti-edematizing potencies of antitoxins prepared by hyperimmunization with each of crude and purified antigens were the same when titrated with either crude or purified lethal toxin as test toxin.
