Personalized therapy for female infertility means determining of immune mechanisms, including allergic sensitization towards sperm antigens. In order to determine antisperm serum IgE antibodies in women with infertility suffering from pelvic inflammatory diseases, the modified Anti-Spermatozoa Antibody protocol was used (ASA Serum ELISA, Demeditec Diagnostisc, Germany). The modification of the protocol is designed to detect only serum antisperm IgE antibodies carrying Fab fragments to sperm antigens.

The distribution of antigenic determinants, recognized by seven anti-carp spermatozoa monoclonal antibodies (MAbs) and two anti-carp spermatogonia MAbs were studied using fresh, unfixed carp spermatozoa. The location of the antigenic determinants was analysed in light- and in scanning electron microscopy, in the latter with the backscattered imaging mode. With all seven anti-carp spermatozoa MAbs a similar regular distribution of the immuno-gold labeling was present on heads, midpieces and tails of carp spermatozoa, whereas with the anti-carp spermatogonia MAbs only low or no labeling was observed.

Sperm (spermatozoa and the various secretions of the accessory glands) with its very complex antigenic system is only produced from puberty and thus at a time when the body has already had a for a long time the facility to respond to an antigenic stimulation with an immune reaction. Because of this, the spermatozoa, in contrast to other cells of the organism, are considered as strangers and may behave as auto-antigens. However, the existence of a haemato-testicular barrier, that prevents all contact between the sperm and the immune system must be recognized in man, its rupture has the consequence of formation of anti-spermatozoa antibodies.
