Using laser light scattering spectroscopy, we are studying age-related changes in the microstructure of lens cytoplasm. We have established in animal models that one of the earliest identifiable stages in cataract development is the presence of a phase transition in the lens cytoplasm. As a result of the phase transition, the cytoplasm separates into microvolumes that differ in their protein concentration. These microvolumes scatter light and cause the lens to become opaque. This phase separation occurs in normal lens cells at a characteristic temperature, Tcat, which varies across the lens with the cell age. As the animal becomes older, the Tcat for the nuclear cells decreases to a value well below body temperature. In X-irradiated eyes, however, Tcat increases with animal age until the phase separation occurs at or near body temperature. At this point, a well-developed nuclear cataract appears. We are now attempting to understand the biochemical basis for the differences between Tcat of normal and Tcat of X-irradiated lens cells during the aging process.

Complex coacervation is a form of liquid-liquid phase separation, whereby two types of macromolecules, usually bearing opposite net charges, self-assemble into dense microdroplets driven by weak molecular interactions. Peptide-based coacervates have recently emerged as promising carriers to deliver large macromolecules (nucleic acids, proteins and complex thereof) inside cells. Thus, it is essential to understand their assembly/disassembly mechanisms at the molecular level in order to tune the thermodynamics of coacervates formation and the kinetics of cargo release upon entering the cell.

The ternary complex effectively prevents droplet aggregation, Ostwald ripening, and phase separation through its gel network, thereby demonstrating its capability in bioactive compound delivery. In this work, the influence of varying chickpea protein isolate (CPI) levels on the microstructure, gel characteristics, stability and functional properties of grape seed proanthocyanidin (GSP) and konjac gum (KGM) stabilized ternary complexes was investigated. Visual appearance indicated the formation of a non-stratified ternary complex as the CPI enhanced to 3-4 %.

Efforts to reduce the impact of chemical processes on the environment are leading to a shift to enzymatic alternatives, with laccases standing out for their versatile substrate oxidation capabilities. This study addresses the improvement of biocatalytic reactions by deep eutectic solvents (DES), in particular DES-based aqueous two-phase systems (ATPS) for the extraction of biomolecules.
