Piracanjuba (Brycon orbignyanus) is an endangered fish species from the Neotropical region. The establishment of a cryobank using spermatogonial stem cells (SSCs) and subsequent production of a germline chimera is thus a promising strategy for such species. In the present work, procedures for the isolation and cryopreservation of piracanjuba SSCs and subsequent transplantation into sterile recipients were established.
View Article and Find Full Text PDFThe biological effect of the extracellular peptide reactivating factor (RF) from Luteococcus casei on cells of probiotic cultures was studied. The RF showed the protective and reactivating effects on the Bifidobacterium bifidum cells under the action of bile salts and an acidic stress. Also, it acted as a cryoprotector during lyophilisation and long-term culture storage.

Some causes of the DNA disorders, the reactive oxygen species, especially, are discussed. View Article and Find Full Text PDFCryoconservation effects on macromolecular parameters of DNA from reproductive cells of fishes are studied as well as cryodamages of DNA in solutions under various freezing conditions, by the viscometry method. A dependence of the low temperature action on DNA concentration in solution and on the action duration is determined. It is stated as well that the nitrogen temperature action on the biopolymer solution results in the formation of both double- and single-stranded breaks.
View Article and Find Full Text PDFA comparative analysis of two types of cryoprotectants, antifreeze glycoproteins from Atlantic cod (Gadus morhua) and carbohydrates from hemolymph of amphypoid crustacean (Gammarus lacustris) has been performed. Both glycoprotein and carbohydrate antifreezes effectively decreased the freezing point of water solutions and diminished the size of ice crystals formed. Noncolligative and colligative mechanisms of action are characteristic correspondingly for glycoproteins and carbohydrates.
