Using a single mini-octadecylsilane (ODS) 5-micron ultrasphere column (0.46 X 4.5 cm) and linear gradients of different solvents, all the aspects of protein structural analysis have been defined. The effectiveness of the system has been evaluated by separating the alpha and beta chains of hemoglobin and their tryptic peptides, then performing amino acid analysis and, finally, identifying the phenylthiohydantoin derivatives of amino acids.

Assessing CYP2E1 phenotype in vivo may be important to predict individual susceptibility to those chemicals, including benzene, which are metabolically activated by this isoenzyme. Chlorzoxazone (CHZ), a specific CYP2E1 substrate, is readily hydroxylated to 6-OH-chlorzoxazone (6-OH-CHZ) by liver CYP2E1 and the metabolic ratio 6-OH-CHZ/CHZ in serum (MR) is a specific and sensitive biomarker of CYP2E1 activity in vivo in humans. We used this MR as a potential biomarker of effect in benzene-treated rats and, also, in humans occupationally exposed to low levels of benzene.

Sorafenib, a new oral multikinase inhibitor with antiangiogenic properties, has demonstrated preclinical and clinical activity against several tumor types. The aims of this study were to validate a method for the measurement of sorafenib in plasma from cancer patients, then to test this method in clinical practice. Following liquid-liquid extraction, the compounds were separated with gradient elution (on a C18 ultrasphere ODS column using a mobile phase of acetonitrile/20 mM ammonium acetate), then detected at 255 nm.

An analytical method for quantitation of colistin A and colistin B in plasma and culture medium is described. After protein precipitation with acetonitrile (ACN) containing 0.1% trifluoroacetic acid (TFA), the supernatants were diluted with 0.

Calculation of the HPLC chromatographic retention times of different carotenoids supported our improved chromatographic separation of beta-carotene and lutein in four tomatoes and two tomatillo varieties in fresh form and after three different cooking procedures: pot boiling, cooking in a pressure cooker and microwaving. A good separation was achieved experimentally using an Ultrasphere ODS column and gradient elution with an acetonitrile-tetrahydrofuran-water mobile phase. It was shown that diverse tomato species contained different amounts of beta-carotene (6-400 microg/100 mg) and lutein (2-30 microg/100 mg).

Domperidone is currently used in Canada and Europe for the treatment of intestinal motility disorders as well as for its antiemetic properties. Recent drug metabolism studies have indicated that domperidone is a substrate of different subtypes of CYP3A family and consequently, the drug requires complete characterization of its metabolism for the identification of major drug-drug interactions. Therefore, the purpose of our studies was to develop a simple, sensitive and rapid HPLC assay for the determination of domperidone and its major metabolites.
