Generalized, sublethal damage-based mathematical approach for improved modeling of clonogenic survival curve flattening upon hyperthermia, radiotherapy, and beyond.

Mathematical modeling can offer valuable insights into the behavior of biological systems upon treatment. Different mathematical models (empirical, semi-empirical, and mechanistic) have been designed to predict the efficacy of either hyperthermia (HT), radiotherapy (RT), or their combination. However, mathematical approaches capable of modeling cell survival from shared general principles for both mono-treatments alone and their co-application are rare.

Positive affect promotes mental health and physical well-being, which may involve modifications in the autonomic nervous system activity. Here, we examine, using chemogenetic techniques, the effects of nucleus accumbens (NAc) activation on affect and body temperature regulation as a proxy of autonomic function. A conditioned place preference test revealed that nucleus accumbens activation induced positive affect.

Introduction: Attention Deficit Hyperactivity Disorder (ADHD), a prevalent neurodevelopmental disorder affecting a significant portion of the population, is commonly managed with stimulant medications. These medications, while effective, have been associated with thermoregulatory dysfunction and an increased risk of heat-related adverse events. The current study sought to compare the incidence of such events in ADHD patients receiving stimulant medications with those not on these treatments.

Hyperthermia has emerged as a popular treatment option due to its high efficacy and seamless integration with other therapeutic approaches. To enhance treatment outcomes, hydrogels loaded with photothermal agents and activated by near-infrared (NIR) light for localized tumor therapy have attracted considerable attention. This approach minimizes drug dosage and mitigates the adverse effects of systemic drug delivery on healthy tissues.

This paper describes the design and initial proof-of-concept of a single pre-clinical transcranial focused ultrasound (FUS) system capable of performing histotripsy (mechanical ablation), hyperthermia, blood-brain barrier opening (BBBO), sonodynamic therapy, or neuromodulation in a murine brain. We have termed it the All-in-One FUS system for murine brain studies, which is the first FUS system of its kind. The 1.
