Pigs on nutritional studies developed heterotopic calcifications in the left atrial endocardium, submucosa of the gastric fundus and to a lesser extent in other sites. Light and electron microscopy of mineralized tissue showed early edema and connective tissue change, deposition of calcium, granulomatous reaction and repair. There was no alkaline phosphatase activity at the sites of calcification and the distribution of this enzyme activity was normal in other tissues. Increased acid phosphatase activity was demonstrated only in the lysosomes of macrophages in areas of granulomatous inflammation. Calcium and magnesium were within normal range in serum and femoral bone. The character and distribution of the lesions suggested the possibility of protracted low grade hypervitaminosis D3.

Oscillatory fluid flow enhanced mineralization of human dental pulp cells.

Front Bioeng Biotechnol

January 2025

Department of Preventive Dentistry, Division of Pediatric Dentistry, Faculty of Dentistry, Naresuan University, Phitsanulok, Thailand.

The purpose of this study is to evaluate the optimum frequency of oscillatory fluid flow (OFF) for increasing osteogenesis in human dental pulp cells (DPCs) in an incubating rocking shaker. DPCs from 3 donors were cultured in an osteogenic induction medium (OIM) and mechanical stimulation was applied using an incubating rocking shaker at frequencies of 0 (control), 10, 20, 30, and 40 round per minute (RPM) for 1 h/day, 5 days/week. Cell proliferation was measured using total protein quantification, and osteogenic activity was measured by alkaline phosphatase (ALP) activity, calcium deposition, and collagen production on days 7, 14, and 21 of culture.

A co-signaling receptor, 2B4, has dual effects in immune cells, but its actual functions in T cells remain elusive. Here, using super-resolution imaging technology with an immunological synapse model, we showed that 2B4 forms "2B4 microclusters" immediately after 2B4-CD48 binding. A lipid phosphatase, SHIP-1, subsequently combined with 2B4 to form coinhibitory signalosomes, leading to the suppression of cytokine production.

Line-Shifting Triarylmethyl Radicals for Imaging of Enzyme Activity Using Overhauser-Enhanced Magnetic Resonance Imaging: Application to Alkaline Phosphatase.

Chem Biomed Imaging

January 2025

In Vivo Multifunctional Magnetic Resonance Center, Robert C. Byrd Health Sciences Center, West Virginia University, Morgantown, West Virginia 26506, United States.

Enzyme catalytic activities are critical biomarkers of tissue states under physiological and pathophysiological conditions. However, the direct measurement and imaging of enzyme activity remains extremely challenging. We report the synthesis and characterization of the first stable triarylmethyl (TAM) radical substrate of alkaline phosphatase (TAM-ALPs).

Ionic silver coating of orthopedic implants may impair osteogenic differentiation and mineralization.

Exp Ther Med

March 2025

OrthoLab, The Rudbeck Laboratory, Department of Surgical Sciences/Orthopedics, Uppsala University, 75185 Uppsala, Sweden.

Silver (Ag) possesses potent antimicrobial properties and is used as a coating for medical devices. The impact of silver ions released from orthopedic implants on the differentiation and osteoid formation of different osteogenic cells has yet to be systematically studied. In the present study, human mesenchymal stem cells (hMSCs) and primary human osteoblasts (hOBs) were exposed to different static Ag concentrations (0, 0.

Bispecific antibodies (BsAb) have emerged as a leading treatment modality in patients suffering from B-cell non-Hodgkin's lymphoma (B-NHL). However, treatment failure is common and may potentially be attributed to pre-existing or emerging T-cell exhaustion. CD39 catalyzes-together with CD73-the hydrolysis of immunogenic ATP into immunosuppressive adenosine and thus actively promotes an immunosuppressive micromilieu.
