Precise prefractionation of proteome samples is a potent method for realizing in-depth analysis in top-down proteomics. PEPPI-MS (Passively Eluting Proteins from Polyacrylamide gels as Intact species for MS), a gel-based sample fractionation method, enables high-resolution proteome fractionation based on molecular weight by highly efficient extraction of proteins from polyacrylamide gels after SDS-PAGE separation. Thereafter it is essential to effectively remove contaminants such as CBB and SDS from the PEPPI fraction prior to mass spectrometry.

Separation of a monoclonal antibody (mAb) from impurities was examined on different cation exchange resins (CEX), including POROS XS, POROS HS, NUVIA S, and NUVIA HRS. Impurities mainly consisted of cell culture-derived mAb fragments, or lysozyme, that mimicked the presence of an adsorbing protein of low molecular weight. The choice between the flowthrough mode and the bind-and-elute mode for the purification was guided by the shape of the adsorption isotherm.

Biosurfactants are biodegradable, non-toxic, and environmentally beneficial substances that are produced by microorganisms. Due to their chemical characteristics and stability in various environmental circumstances, biosurfactants are low-molecular-weight, surface-active molecules of great industrial importance. The choice of the producer microbe, kind of substrate, and purification technique determine the chemistry of a biosurfactant and its production cost.

Bispecific antibodies hold significant potential as next-generation biotherapeutics owing to their ability to simultaneously bind to two targets. However, the development of bispecific antibodies as biotherapeutics has been hindered by the high levels of byproducts produced, including both high molecular weight and low molecular weight variants. In addition, the inevitable expression of homodimers in host cells presents further obstacles to the commercial development of bispecific antibodies as therapeutics.
